Description of key information
Preliminary test showed that it could be reasonable to expect that estimated EC50 will be higher than solubility of the test substance. Water solubility of the test substance is 0.0746 mg.L-1, and the average determined concentration in M4 medium was 0.0539 mg.L-1.

When the assumed value of EC50 is higher than solubility of the test substance in M4 medium, no further testing is needed and the result obtained in preliminary test presents the result of whole study.

The test substance, Anthraquinone, was tested in reproduction test on Daphnia magna according to submitted testing proposal.
The test was performed according to method C.20 – Daphnia magna Reproduction Test, Council Regulation (EC) No. 440/2008, published in O.J.L 142, 2008.
Since the test substance is poorly soluble in M4 medium at concentrations needed for the test, the saturated solution of the test substance in M4 medium was prepared for preliminary test. The saturated solution was prepared by adding 1 mg.L-1of the test substance to M4 medium. The saturated solution was stirred for period 72 hours on a shaft stirrer and consequently filtered through 0.45 μm filter.
The preliminary test was performed in range of test substance concentration 1000 – 125 mL.L-1. The test was performed as semi-static, frequency of the test solutions renewal was three times a week. Samples for analytical determination were taken at each test concentration several times during of the test (before and after renewal of the test solutions).
The highest calculated reduction in reproductive output was 1.8 %, the exact values of EC50 could not be calculated on the basis of data acquired. Test result is given in the form of a range.
For all evaluations and results the measured concentration of the test substance were used.
