Additional information

In a primary study the skin irritation potential of 1 -chloro-2 -nitrobenzene was investigated comparable to OECD guideline 404 with acceptable restrictions. An 2x2 cm area of an albino rabbit ear inside was covered 24 h to applicate the test substance. Cellulose pads were loaded with 500 mg 1 -chloro-2 -nitrobenzene. The test areas were covered semi-occlusive with a layer of cellulose and plaster. No skin irritating effects were observed throughout the test and after 7 days observation time (Groening, Bayer AG, 1976).

The skin irritation potential of 1 -chloro-2 -nitrobenzene was also evaluated in a study performed comparable to OECD guideline 404. The intact and abraded skin flank of six Himalayan rabbits was exposed to 0.5 ml 10% (dissolved in sesame oil) test material for 24 hours under semi-occlusive conditions. The rabbits were observed for 72 hours. Skin reactions were assessed using the Draize scheme according to Fed.Reg.38, No.187, p.27019, 1973, § 1500.41 after 24, 48 and 72 hours after substance application. The mean erythema scores after 24 hours were 1.0 for both the intact and abraded skin in 4/6 animals. The edema scores were 0.0 for the intact and abraded skin after 24, 48 and 72 hours. All treated skin sites appeared normal at the 48-hour observation (Hoechst AG, 1975).

The eye irritation potential of 1 -chloro-2 -nitrobenzene was investigated in 6 Himalyan rabbits according to the FDA guideline (federal register, Vol 38, No. 187, 1973). 100 mg undissolved test substance was applied into the conjunctival sac of one eye of each animal. The eyes were rinsed after 24 hours application of the test substance. The eye irritation reactions were examined and scored using the Draize method 1, 7 and 24h after application. After 1 hour 1 -chloro-2 -nitrobenzene induced slight conjunctival redness in all animals (4/6 score=1 and 2/6 score=2). These effects were reversible within 24 h. The mean scores over 1, 7 and 24 h were 0.0 for corneal opacity, iritis, chemosis and conjunctivae in all animals. Therefore no classification is required (Hoechst AG, 1975).

In another study comparable to OECD guideline 405 the eye irritation potential of 1 -chloro-2 -nitrobenzene was investigated in 2 albino rabbits. 50 mg test substance was applied into the conjunctival sac of the right eye of each animal. No eye irritating effects (conjunctivae and cornea) were observed throughout the test and after 7 days observation time (Groening, Bayer AG, 1975).

Based on the study results and according to EU classification criteria, the test substance is not to be classified.
