Two hydrolysis tests were performed according to test guideline with substance with ELINCS No. 405-370-0, which is structurally similar to N,N,N',N'-tetrakis(2-hydroxypropyl)adipamide (EINECS No. 260-982-3).


One test (David A. Howes, 1990) was performed according to EU method C.7 to evaluate the hydrolysis of the test item as a function of pH. The test item was dissolved in aqueous buffer solution, and the temperature and pH were checked at pH 4, 7 and 9, respectively. The decrease of the concentration with time was determined by HPLC. The rate constant and the half-life period were calculated using the slope of the line (the logarithms of the concentration against time). The test results showed that the test substance possesses a half-life of greater than 1 year at 25 °C and pH 4, approximately 1 year at 25 °C and pH 7, and less than 1 year at 25 °C and pH 9.


Another test (Ian A. Macdonald, 1991) was performed according to EU method C.7 for further test at pH 9. The hydrolysis of the test item was checked at the temperature of 50 °C, 60 °C and 70 °C at pH 9. The decrease of the concentration with time was determined by HPLC. The rate constant and the half-life period were calculated using the slope of the line (the logarithms of the concentration against time). The test results showed that the half-life of QM-552 at pH 9.0 and at 25 °C is 2725 hours (113.54 days) based on the (pseudo-) first order reaction.
