Additional information

Only very slight edema and erythema were observed in a test for skin irritation of 3,3'-dichlorobenzidine dihydrochloride in rabbits which were exposed for 1 or 4 hours. These effects were fully reversible within the 72 hour post exposure observation period. These findings were supported by two other studies of less reliability, one performed with the submission substance and one performed with the free base 3,3'-dichlorobenzidine. Therefore, the submission substance has not to be classified for skin irritation according to Regulation (EC) No 1272/2008 and Council Directive 67/548/EEC.

3,3'-Dichlorobenzidine dihydrochloride caused severe irritation to the eyes after application of a 20% suspension in corn oil into the conjunctival sac of the rabbit eye. However, these data were judged not to be reliable due to insufficient reporting. But, due to the strong acid reaction of an aqueous solution of 3,3'-dichlorobenzidine dihydrochloride (the pH of a 1% aqueous solution is <2), it can reasonably be assumed, that contact of the test substance with the mucous membranes of the eye results in the formation of a strong acid slurry, which causes severe burns to eyes (see OECD TG 405). Therefore, 3,3'-dichlorobenzidine dihydrochloride should be classified as substance which "Causes serious eye damage" (H318; eye damage cat. 1) according to Regulation (EC) No 1272/2008 or as substance with "Risk of serious damage of eyes" (R41) according to Council Directive 67/548/EEC without further testing.

Justification for classification or non-classification

3,3'-Dichlorobenzidine dihydrochloride is not irritating to skin and has not to be classified for skin irritation according to Regulation (EC) No 1272/2008 and Council Directive 67/548/EEC.

3,3'-Dichlorobenzidine dihydrochloride causes severe burns to rabbit eyes. Additionally, an aqueous slurry of the test item has a strong acid pH of <2. Therefore, 3,3'-dichlorobenzidine dihydrochloride should be classified as substance which "Causes serious eye damage" (H318; eye damage cat. 1) according to Regulation (EC) No 1272/2008 or as substance with "Risk of serious damage of eyes" (R41) according to Council Directive 67/548/EEC.
