The aquatic toxicity of the mixed-Tocopherols (CAS: 1406-66-2) is evaluated on the basis of read across data to DL-alpha-Tocopherol (CAS: 10191-41-0) and/or of the precursor of alpha-Tocopheryl acetate (CAS: 7695-91-2).

acute toxicity in fish:

The acute toxicity in fish was evaluated on the basis of a read across data to the structural isomer DL-alpha-tocopherol (CAS: 10191-41-0): The test was performed according to OECD 203 in rainbow trout (Oncorhynchus mykiss(reported asSalmo gairdneri))under GLP. After 96 h exposure a LC0/LC50 were determined to be > 10 mg/L.

  

acute toxicity in daphnia:

The acute toxicity in daphnia was evaluated on the basis of a read across data to the structural isomer DL-alpha-tocopherol (CAS: 10191-41-0) and /or the precursor DL-alpha-tocopheryl acetate (CAS: 7695-91-2):

 

One test was performed with DL-alpha-tocopherol (CAS: 10191-41-0) according to OECD 202 in waterflea (Daphnia magna) under GLP. After 48 h exposure the EC 50 was determined to be > 23 mg/L .

 

A further test was performed for DL-alpha-tocopheryl acetate (CAS: 7695-91-2) according to DIN 38412/11 without GLP, in waterflea (Daphnia magna). In the study the EC 50 after 48 h exposure was determined to be > 500 mg/L .

 

 

 acute toxicity in algae:

The acute toxicity in algae was evaluated on the basis of a read acorss data to the structural isomer DL-alpha-tocopherol (CAS: 10191-41-0) and/or the precursor DL-alpha-tocopheryl acetate (CAS: 7695-91-2):

 

One test was performed with DL-alpha-tocopherol (CAS: 10191-41-0) according to OECD 201 and GLP. After 72 h exposure the EC 50 and NOEC were determined to beto be > 25.8 mg/L and 25.8 mg/L, respectively,related to growth rate.

 

A second test was performed for DL-alpha-tocopheryl acetate (CAS: 7695-91-2) according to OECD 201 and GLP. After 72 h exposure the EC 50 and NOEC were determinedto be > 27.8 mg/L and 27.8 mg/L, respectively,related to growth rate.

 

acute toxicity against mircoorganisms:

Two bacteria tests for read across are available for the group of tocopherols and tocopheryl acetates:

One test was performed with DL-Alpha-Tocopheryl acetate, a precursor of the substance. The respiration rate of microorganisms in presence of DL-Alpha-Tocopheryl acetate was tested by a oxygen consumption test according to ROBRA focusing on the respiration of Pseudomonas putida. After 30 minutes an EC10 > 10000 mg/l was determined (BASF AG, 1991).

In another test changes in the respiration rate of activated sludge as induced by DL-Alpha-Tocopheryl acetate were studied in a test performed in compliance with ISO 8192. An EC20 > 927 mg/l was determined after an inoculation time of 30 minutes.

The study with Pseudomonas putida (BASF AG, 1991) is selected as key study.
