Hexylene glycol was reported to have low acute toxicity to various fish species with 96-hour LC50 values well above 5000 mg/L. In Gambusia affinis, the 96-hour LC50 of hexylene glycol was determined to be 8510 mg/L. LC50 values within the > 5000 to 13 500 mg/L range also were reported for Pimephales promelas, Lepomis macrochirus, Oncorhynchus mykiss, Carassius auratus affinis, Ictalurus punctatus, Alburnus alburnus, and Menidia beryllina following 96 hours of exposure to hexylene glycol.

 

Hexylene glycol also was reported to have low acute toxicity to various aquatic invertebrate species. The 48-hour EC50 of hexylene glycol was determined to be 5410 mg/L in Daphnia magna, 3300 mg/L in Daphnia pulex, and 2800 mg/L in Ceriodaphnia sp. Other endpoints measured included mortality, inhibition of growth, and immobility, the results of which support the low acute toxicity of hexylene glycol in Daphnia magna, Tanytarsus dissimilis, Orconectes immuni, Arbacia punctulata, Nitocra spinipes, and Artemia salina.

Hexylene glycol was reported to have low toxicity to Selenastrum capricornutum (i.e.,Pseudokirchnerella subcapitata), with a 72-hour NOEC of 429 mg/L. The 72-hour EC50 values for both biomass (EbC50) and growth rate (ErC50), as well as the 72-hour EC10 values for these endpoints, were determined to be greater than 429 mg/L. Therefore, hexylene glycol did not exhibit growth inhibiting properties when tested in algae.

Hexylene glycol was determined to have low toxicity in microorganisms. Specifically, an EC0 (equivalent to a NOEC) of 200 mg/L was determined in Pseudomonas aeruginosa. Supporting data from a luminescence study in Photobacterium phosphoreum indicate an EC50 of 3070 mg/L. In addtion, the LC50 of hexylene glycol to tadpoles (Rana catesbeiana) was reported to be 11800 mg/L.
