One study investigating the toxicity of 1,2,3-Propanetriol, homopolymer (CAS 25618-55-7) to aquatic microorganisms is available (Clariant, 1989). In addition, in accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5, a read across to the two structurally related analogue substances Reaction mass of 3-[3-(2,3-dihydroxypropoxy)-2-hydroxypropoxy]propane-1,2-diol, 3-(2,3-dihydroxypropoxy) propane-1,2-diol,3-[3-[3- (2,3-dihydroxypropoxy)-2-hydroxypropoxy]-2-hydroxypropoxy]propane-1,2-diol (List No. 915-741-3) and glycerol (CAS 56-81-5) was conducted, since the own study is not considered to be sufficient to cover the endpoint, due to the poor documentation of the experimental details.

This read-across is justified in detail in the overall endpoint summary in IUCLID Section 6.1. Both read-across substances represent main components of the target substance (mono- to tetraglycerol). Thus, the aquatic toxicity pattern given by these read across substances are considered to be adequately assessing the aquatic toxicity pattern of 1,2,3-Propanetriol, homopolymer (CAS 25618-55-7).

Together, the own study and the read across studies cover the endpoint in a “weight-of-evidence” approach.

The first study with 1,2,3-Propanetriol, homopolymer (CAS 25618-55-7) was performed according to DIN guideline 38412, part 8 under static conditions using Pseudomonas putida as test organism (Hoechst, 1989). The study was not performed under GLP conditions. No adverse effects were observed up to a test concentration of 1000 mg/L. Thus, the EC10 is determined to be > 1000 mg/L based on the nominal concentration. 

The second study investigated the toxicity of the read across substance Reaction mass of 3-[3-(2,3-dihydroxypropoxy)-2-hydroxypropoxy]propane-1,2-diol, 3-(2,3-dihydroxypropoxy)propane-1,2-diol,3-[3-[3-(2,3-dihydroxypropoxy)-2-hydroxypropoxy]-2-hydroxypropoxy]propane-1,2-diol (List No. 915-741-3) to aquatic microorganisms (Desmares-Koopmans, 2009). The test was performed according to the OECD Guideline 209 under GLP conditions usingactivated sludge of a predominantly domestic sewageas test organism. A limit test with a test substance concentration of 100 mg/L was applied. As no significant inhibition of the respiration rate of the sludge was recorded after an exposure time of 3 h the NOEC value is determined to be >= 100 mg/L based on the nominal concentration.

A third study with read-across substance glycerol (CAS 56-81-5) was performed according to DIN guideline 38412, part 8 under GLP conditions using Pseudomonas putida as test organism (Henkel, 1994). No cell growth inhibition was measured in any treatment after an exposure time of 16 h. Thus, the EC10 is determined to be > 10000 mg/L based on nominal test concentrations.

The own study and the studies with the read-across substances, composed of main components of the target substance (mono-, polyglycerols), consistently show no adverse effects up to the highest concentration tested. Thus, it can be concluded that 1,2,3-Propanetriol, homopolymer (CAS 25618-55-7) will not exhibit inhibitory effects on aquatic microorganisms.
