1 (reliable without restriction)
Rationale for reliability incl. deficiencies:
guideline study
Qualifier:
according to guideline
Guideline:
OECD Guideline 203 (Fish, Acute Toxicity Test)
GLP compliance:
yes (incl. QA statement)
Analytical monitoring:
yes
Details on sampling:
- concentrations - all
- Sampling method: 100 uL taken and added to appropriate vials to maintain conconcentation
- Sample storage conditions before analysis: stored at 2-10 cegrees Celcius and shipped to another facility at the end of the test (stability during storage and transport shown in another study)
Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
Vial filled with test solutions that had been purged for 1 hour with the test material at a rate of approximately 10 mL/min. The test vessels were filled to the rim to minimize headspace and closed with transparent lids sealed with a silicone vacuum grease. This method was shown in another experiment to result in a saturated solution.
Test organisms (species):
Cyprinus carpio
Details on test organisms:
TEST ORGANISM
- Common name: carp
- Source: Zodiac, LU Wageningen, the Netherlands
- Length at study initiation : 2.4-2.8 cm
- Weight at study initiation (mean and range, SD): 0.49 +/- 0.08g
- Method of breeding: Fi from a single parent-pair bred in UV treated water
- Feeding during test - no

ACCLIMATION
- Acclimation period: at least 12 days
- Acclimation conditions (same as test )
- Type and amount of food: pelleted fish food
- Feeding frequency: daily
- Health during acclimation (any mortality observed): no
Test type:
static
Water media type:
freshwater
Limit test:
yes
Total exposure duration:
96 h
Test temperature:
Day 0 - 21.1-21.2C
Day 4 - 21.5-21.3C
pH:
Day 0 - 7.9 - 8.2
Day 4 - 7.3-7.2
Dissolved oxygen:
Day 0 - 6.4 - 9.2
Day 4 - 2.7 - 3.2
Nominal and measured concentrations:
nominal - above 100 mg/L
measured - 197 mg/L
Details on test conditions:
TEST SYSTEM
- Test vessel: 10 liter, all glass closed with transparent lids sealed with silicone high vacuum grease
- Type : closed
- fill volume:9.8 liters
- Aeration:no
- No. of organisms per vessel:7
- No. of vessels per concentration (replicates): 2
- No. of vessels per control (replicates): 1
- Biomass loading rate: 0.15 g fish/liter

OTHER TEST CONDITIONS
- Adjustment of pH: no
- Photoperiod: 16 hours daily

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) :

TEST CONCENTRATIONS
- Spacing factor for test concentrations:Not applicable, limit test
- Test concentrations: 100 mg/L
Reference substance (positive control):
yes
Remarks:
pentachlorophenol
Key result
Duration:
96 h
Dose descriptor:
LC50
Effect conc.:
> 197 mg/L
Nominal / measured:
meas. (arithm. mean)
Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
- Behavioural abnormalities:
- Mortality of control: none
- Other adverse effects control:no
Validity criteria fulfilled:
yes
Remarks:
no mortality in control, test conditions maintained during test, dissolved oxygen content at least 60% air saturation value, results based on geometric average measured concentrations
Conclusions:
The 96 hour LC50 for carp exposed to the test substance is > 197 mg/l and the test substance can be classified as not harmful to freshwater fish
