Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Additional information

There were no data available for the registered substances so key studies for the related substances, chloro(dimethyl)silane (CAS number 1066-35-9 ) and Dichloro(dimethyl)silane (CAS number 75 -78 -5) were selected as key studies. A bacterial mutagenicity study on the related substance hexamethyldisilane (CAS number 1450 -14 -2) has also been taken into consideration. The results of all the studies were in agreement. It is considered appropriate to read across to the registered substance which is a mixture of chloromethyl disilanes the genetic toxicity results for chloro(dimethyl)silane and dichloro(dimethyl)silane as they are considered close structural analogues of chloro(methyl)silane: all are small molecule alkylchlorosilanes containing Me-Si and Cl-Si groups, and hexamethyldisilane contains a Si-Si group. In addition, bacterial mutagenicity results for the read-across substances are in agreement, and none of the substances contains a structural alert for genetic toxicity.

Short description of key information:
In vitro:
Gene mutation (Bacterial reverse mutation assay / Ames test): read-across from analogous substance CAS 1066-35-9 negative with and without activation in all strains tested (OECD TG 471)
Gene mutation (Bacterial reverse mutation assay / Ames test): read-across from analogous substance CAS 75-78-2 negative with and without activation in all strains tested (similar to OECD TG 471)
Gene mutation (Bacterial reverse mutation assay / Ames test): read-across from analogous substance CAS 1450-14-2: negative with and without activation in strains TA98, TA100, TA1535 and TA 537

Cytogenicity in mammalian cells: read-across from analogous substance CAS 75-78-2: negative in L5178Y mouse lymphoma cells (similar to OECD TG 473)
Mutagenicity in mammalian cells: read-across from analogous substance CAS 75-78-2: negative in L5178Y mouse lymphoma cells (similar to OECD TG 476)

In vivo:
Chromosome aberration assay in rat (ip administration): read-across from analogous substance CAS 75-78-2: negative in bone marrow cells (similar to OECD 475)

Endpoint Conclusion: No adverse effect observed (negative)

Justification for classification or non-classification

There is no information available fro chloro(methyl)silane. The analogous substances chlorodimethylsilane (CAS number 1066 -35 -9), dichloro(dimethyl)silane (CAS number 75 -78 -5) and hexamethyldisilane (CAS number 1450-14-2) do not cause mutagenicity in bacteria. Data is available for dichloro(dimethyl)silane (CAS number 75 -78 -5) for in vitro mammalian mutagenicity and cytogenicity and an in vivo chromosome aberration assay, and these negative results can be read across to the registration substance. Therefore it is considered that classification for mutagenicity is not required.
