Principles of method if other than guideline:
To evaluate the applicability of an in vivo-in vitro replicative DNA synthesis (RDS) test using rat hepatocytes, we conducted the RDS test with 22 nongenotoxic (Ames-negative) hepatocarcinogens and 25 noncarcinogens (including 3-chloro-p-toluidine, CAS 95-74-9) under standardized conditions and judgement criteria.

Applicant's summary and conclusion

Executive summary:

3-chloro-p-toluidine clearly stimulated RDS at 24 or 39 h after treatment. 3-chloro-p-toluidine is regarded as a noncarcinogen. Therefore the test system is not suitable for mutagenicity testing of 3-chloro-p-toluidine.
