Description of key information

Aquatic toxicity of metformin HCl

Study Type
Organism
Effect concentation
Acute toxicity (TAD 4.08)
Daphnia magna 48h-NOEC: 78 mg/L;48h-EC50: 130 mg/L
Acute toxicity (EU C.2)
Daphnia magna 48h-EC50: 64 mg/L (literature study, supportive information)
Acute toxicity (TAD 4.11) Lepomis macrochirus 96h-NOEC: 982 mg/L; 96h-LC50: >982 mg/L
Chronic toxicity (OECD 211) Daphnia magna 21d-NOEC:  32 mg/L
Chronic/Acute toxicity Algae (EU C.3) Desmodesmus subspicatus 72h-EC50: >320 mg/L
Chronic toxicity (ISO /WD20079) Lemna minor 7d-EC50: 110 mg/L
Chronic toxicity (OECD 210) Danio rerio 34d-NOEC: ≥12 mg/L

In general, metformin HCl has a very low hazardous potential on aquatic organisms. In acute aquatic toxicity tests, metformin HCl did not cause any adverse effects to fish (Lepomis macrochirus) and only weak effects to invertebrates (Daphnia magna). Chronic toxicity testing with aquatic organisms also resulted in either no (Desmodesmus subspicatus, Daphnia magna, Danio rerio) or weak (Lemna minor) adverse effects at the concentrations tested. The lowest NOEC from aquatic toxicity studies was the 34d-NOEC of  12 mg/L obtained in the Early life stage test with Danio rerio.

Aspergillus clavatus MIC: >1000 mg/L
Penicillium canescens MIC: >1000 mg/L
Chaetomium globosum MIC: >1000 mg/L
Pseudomonas fluorescens MIC: >1000 mg/L
Anabaena flos-aquae MIC: 100 mg/L
