Biodegradation in water: screening tests

Various study for the test compound4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid(CAS No. 119-40-4) and for its read across substance were reviewed for the biodegradation end point which are summarized as below:

Biodegradation study was conducted for 28 days for evaluating the percentage biodegradability of test substance 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid (CAS no. 119 -40 -4). Concentration of inoculum i.e, sludge used was 30 mg/l and initial test substance conc. used in the study was 100 mg/l. Aniline was used as a reference substance for the biodegradation study. In assessment of the Ready Biodegradability of the substance7-anilino-4-hydroxynaphthalene-2-sulphonic acid, the result for degradation were evaluated to be 0%,2% and 0% degradation after 28 days based on O2consumption, DOC removal and HPLC parameter respectively. Therefore, the substance 7 -anilino-4 -hydroxynaphthalene-2- sulphonic acid is determined to be non-biodegradable in water under the test conditions.

Estimation Programs Interface Suite (EPI suite, 2016) was run to predict the biodegradation potential of the test compound 4-Hydroxy-7 -

(phenylamino) naphthalene-2-sulfonic acid (CAS no. 119- 40 -4) in the presence of mixed populations of environmental microorganisms. The biodegradability of the substance was calculated using seven different models such as Linear Model, Non-Linear Model, Ultimate Biodegradation Timeframe, Primary Biodegradation Timeframe, MITI LInear Model, MITI Non-Linear Model and Anaerobic Model (called as Biowin 1-7, respectively) of the BIOWIN v4.10 software. The results indicate that 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid is not expected to be readily biodegradable.

Biodegradation study of substance 2-Amino-8-naphthol-6-sulfonic acid in water was carried out for 14 days(J-CHECK, 2016). The test substance is found to be 5.3 % biodegraded in water after 2 wk on the basis of O2 consumption, BOD. 0.4% by TOC removal and <0 % by (UVVIS) parameters. Hence, the 2-Amino-8-naphthol-6-sulfonic acid is considered to be not biodegradable in water under the test conditions.

A closed bottle test according to OECD 301D guidelines was performed to determine the ready biodegradability of 7-Amino-4 -hydroxy-2 -naphthale nesulphonic acid (GREIM H et. al, 1994). No degradation of test chemical 7-Amino-4-hydroxy-2-naphthalenesulphonic acid was observed. Therefore, the chemical was considered to be not readily biodegradable.

Biodegradation screening test was conducted for 14 days (2 weeks) for evaluating the percentage biodegradability of the test substance(J-CHECK, 2016). Concentration of inoculum i.e, sludge is 30 mg/l and initial test substance conc. used in the study is 100 mg/l. The percentage degradation of test substance is found to be 6.3% by BOD, 8.4% by TOC and 0% by UV-VIS Thus, the substance 7-Amino-4 hydroxy- 2 - naphthalenesulphonic acid was considered to be not readily biodegradable in water.

 

Biodegradation study was conducted for 14 days for evaluating the percentage biodegradability of test substance Sodium naphthol sulfonate (J-CHECK, 2016). Concentration of inoculum i.e, sludge used was 30 mg/l and initial test substance conc. used in the study was 100 mg/l. The percentage degradation of test substance was determined to be 0% by BOD, 2.1% by TOC removal and 1% by UV-Vis in 14 days. Thus, the substance Sodium naphthol sulfonate is considered to be non-biodegradable under the test conditions.

Biodegradation study was conducted for 14 days for evaluating the percentage biodegradability of test substance Naphthol AS (J-CHECK, 2016). Concentration of inoculum i.e, sludge used was 30 mg/l and initial test substance conc. used in the study was 100 mg/l. The percentage degradation of test substance was determined to be 5.4% by BOD in 14 days. Thus, the substance Naphthol AS is considered to be non-biodegradable under the test conditions.

Biodegradation study was conducted for evaluating the percentage biodegradability of test substance Dipotassium 2-naphthol-6,8-disulfonate according to OECD Guideline 301 D (Ready Biodegradability: Closed Bottle Test) (J-CHECK, 2016). No degradation of test chemical Dipotassium

2-naphthol-6,8-disulfonate was observed. Thus, the substance Dipotassium 2-naphthol-6,8-disulfonate is considered to be non-biodegradable under the test conditions.

On the basis of above results for target and read across substance, it can be concluded that the test substance compound 4 -hydroxy-7 -(phenylam ino) naphthalene-2-sulfonic acidcan be expected to be not readily biodegradable in nature.

Biodegradation in water and sediment: simulation tests

Estimation Programs Interface (EPI) Suite (2016) prediction model was run to predict the half-life in water and sediment for the test compound 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid(CAS No. 119 -40 -4). If released in to the environment, 11.2 % of the chemical will partition into water according to the Mackay fugacity model level III and the half-life period of 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acidin water is estimated to be 37.5 days (900 hrs). The half-life (37.5 days estimated by EPI suite) indicates that the exposure risk to aquatic animals is moderate to low whereas the half-life period of 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acidin sediment is estimated to be 337.5 days (8100 hrs). However, as the percentage release of test chemical into the sediment is less than 5% (i.e, reported as 1.67%), indicates that

4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acidis not persistent in nature.

Biodegradation in soil

The half-life period of 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid(CAS No. 119 -40 -4) in soil was estimated using Level III Fugacity Model by EPI Suite version 4.1 estimation database (EPI suite, 2016). If released into the environment, 87.1% of the chemical will partition into soil according to the Mackay fugacity model level III. The half-life period of4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid in soil is estimated to be 75 days(1800 hrs).Based on this half-life value of 4-hydroxy-7 -(phenylamino)naphthalene-2 -sulfonic acid, it is concluded that the chemical is

not persistent in the soil environment and the exposure risk to soil dwelling animals is moderate to low. 

On the basis of available information, the test substance 4-Hydroxy-7-(phenylamino) naphthalene-2-sulfonic acid can be considered to be non-biodegradable in nature
