Acute toxicity to fish

A study was carried out according to EU Method C.1 and OECD Guideline 203 (Fish, Acute Toxicity Test). The 96 hour LC50 for guppy exposed to Nicotinamide was > 1000 mg/L and the NOEC was 1000 mg/L (limit concentration).

Acute toxicity to invertebrates

A study was carried out according to EU Method C.2 and OECD Guideline 202 (Daphnia sp. Acute Immobilisation Test). The 24 hour EC50 was > 1000 mg/L and the NOEC 1000 mg/L (limit concentration).

Acute toxicity to algae

A study was carried out according to EU Method C.3 and OECD Guideline 201 (Alga, Growth Inhibition Test). Scenedesmus subspicatus, fresh water algal growth inhibition test with Nicotinamide. The 72-hour EC50 of growth inhibition and biomass was beyond the range of test concentrations applied in this test (> 1000 mg/L). The no observed effect concentration (NOEC) was 560 mg/L.

Long term aquatic toxicity

According to REACH Annex IX, column 2, section 9.1.6 and 9.1.5 experimental determination of long term toxicity to fish and aquatic invertebrates were waived, as the substance is considered not hazardous to the environment. There is no need to investigate further the effects on aquatic organisms.

Toxicity to microorganisms

A study was carried out similar or equivalent to EU Method C.11 and OECD Guideline 209 (Activated Sludge, Respiration Inhibition Test), specifically following the Umweltbundesamt (UBA) Guidelines: Bewertung wassergefahrdender Stoffe, III Bestimmung der akuten Bakterientoxizitat, Ad-hoc-Arbeidsgruppe I (Obmann Dr. Niemitz), LTwS, Nr. 10, September 1979.

Nicotinamide was investigated for its ability to inhibit the cell multiplication of the bacteria species Pseudomonas putida. Cultures of Pseudomonas putida bacteria were exposed to concentrations ranging from 3.9 to 8000 mg Nicotinamide per liter. After incubation the extinction values of the bacterial cell suspensions were measured and a toxicity threshold value of Nicotinamide of 4235 mg/L for Pseudomonas putida was determined.
