PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Loading rates of 0.160, 0.340, 0.800, 1.70, 3.60 and 8.00 mg/L were prepared by stirring (via magnetic stirrer) in the dark at 700 rpm for 72 h. The media were then filtered by using a 0.45 μm nitrocellulose membrane. The filtrates were stored at room temperature.The filtrate was directly used as the test solution.

A semi-static test design according to OECD 211 was chosen in a study according to Chinese GLP to evaluate the chronic toxicity of aluminium magnesium vanadium oxide to freshwater invertebrates at nominal concentrations from 0.16 mg/L to 8.00 mg/L (2013). The substance has a low water solubility and therefore the test solution was prepared by adding the appropriate test item to the dilution water. After stirring for 72 hours the resulting solution was filtrated through 0.45 µm filters. Following this, the filtrates were used for the main test without dilution. The test solutions included the dilution water control and nominal concentrations of 0.160, 0.340, 0.800, 1.70, 3.60 and 8.00 mg/L test item. Each treatment concentration was prepared separately. The average recoveries of 0.0200 mg/L and 0.160 mg/L Al-spiked samples were 108% and 94.3% with RSDs of 6.46% and 1.29%, and the average recoveries of 0.0200 mg/L and 0.160 mg/L V-spiked samples were 97.3% and 96.8% with RSDs of 0.28% and 1.05%, and the average recoveries of 15 mg/L and 30 mg/L Mg-spiked samples were 106% and 91.9% with RSDs of 5.99% and 5.02%.

The 21 days NOELR was determined to be 0.160 mg/L (corresponding to the time-weighted mean measured concentrations of Al, V and Mg of 0.00833, 0.00213 and 8.64 mg/L). The 21 days LOELR value was determined to be 0.340 mg/L (corresponding to the time-weighted mean measured concentrations of Al, V and Mg of 0.00821, 0.00320 and 8.48 mg/L). The 21 days EL50 value was 3.71 mg/L (95% confidence limits: 1.83-54.4 mg/L).
