Preparation of test solution
-Preliminary test (solution test): prior to range-finding and limit test, solubility test was conducted to identify the solubility of test in dilution water (M4 medium). As a result of solubility test, it's identified that the test substance is soluble with dilution water (M4 medium) at the concentration of 500 mg/L.
-Range finding and limit test: the range finding and limit test are conducted at the nominal concen tration of 27.908, 33.490, 40.188, 48.225, 57.870, 69.444 and 100 mg/L. Each test solution of nominal concentration was prepared by dil ution of stock solution in the morning of exposure date.
-Definitive test: the definitive test be conducted at the nominal concentration of 100 mg/L on the basis of range finding and limit test. Each test solution of nominal concentration was prepared by dilution of stock solution in the morning of exposure date.

TEST method: analysis of the test substance in the range-finding and limit test was carried out by preliminary test according to the SOP for the stability test. As the results of stability test, the concentration changes of test sub stance in nominal concentration (27.908 and 100 mg/L) after 48h were 3.2 and -1.2%, respectively. On the basis of the stability, the definitive test was carried out using static-system.

- Selection of test species: Young daphnids for test were derivied from healthy brood stock of Daphnia magna, deliverd more than three times. At the start of the test, they were aged less than 24 hours.

Immobilization and abnormal behavior: there were no immobility and abnormal behavior in the negative control and nominal concentration of 100 mg/L.

Appearance: all the test solution were kept clear from the beginning to the end of exposure.

Executive summary:

The study was performed to assess the acute toxicity effect of test item to Daphnia magna according to the OECD Guideline for Testing of Chemicals No. 202 (2004).

 

1)      The definitive test was carried out using static system on the basis of stability test.

2)      During the test period, temperature, pH and dissolved oxygen of the test solution were 19.7 ~ 20 °C, 7.99 ~ 8.10 and 7.89 ~ 7.98 mg/L, respectively.

3)      Hardness and Alkalinity of test solution were 228 ~ 240 mg/L (as CaCO3) and 48 ~ 50 mg/L.

4)      All the test solutions were kept clear from the beginning to the end of test.

5)      The geometric mean of measured concentration for test item in the nominal concentration (100 mg/L) was 99.999 mg/L.

 

The results of acute toxicity for test item, the 24h and 48h EC50 of test item based on nominal concentration were great than 100 mg/L, the 24h and 48h EC50 of test item based on measured concentration were great than 99.999 mg/L.
