HPLC method

Details on study design: HPLC method:
EQUIPMENT
- Apparatus: A HPLC comprising of HPLC pump, Injection valve fitted with 100ul loop, 250 x 4.6mm column packed with a 5 um CN phase, PDA scan 210nm to 400nm and chromotography data system.

MOBILE PHASES
- Type: 55% methanol and 45% deionised water pumped through at a flow rate of 1.5 ml/min
- pH: pH range 5.5 to 7.5

DETERMINATION OF DEAD TIME
- Method: by inert substances which are not retained by the column (sodium nitrate)

REFERENCE SUBSTANCES
- Identity: Acetanilide, Atrazine, Isoproturon, Linuron, Naphthalene, 1,2,3-Trichlorobenzene, Fenthion, Phenanthrene, ppDDT

EVALUATION
- Calculation of capacity factors k': k'= (retention time of test substance - column dead time)/ column dead time
- Calculation of retention times: All the reference substances were injected in duplicate, and the mean retention times were determined, Each concentration of test material (5.0 and 10 mg/L) was also injected in duplicate and the mean retention time determined for each concentration.
- Determination of the log Koc value: Determined from the equation of the line from the calibration graph of logKoc versus logk'

Batch equilibrium or other method

Analytical monitoring:
no

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
The log10k' for the test substance could not be calculated at either test concentration (5.0mg/L and 10mg/L) because the peak eluted before the sodium nitrate peak, therefore, it is outside the range and accordingly the estimated log10Koc for the test material is reported as <1.5.
Executive summary:

Introduction

This study was performed to estimate the adsorption coefficient (Koc) of the test material using a high performance liquid chromotography (HPLC) correlation technique. The method was designed to be compatible with:

-OECD Test Guideline 121, Estimation of the Adsorption Coefficient (Koc) on Soil and on Sewage Sludge using HIgh Performance Liquid Chromotography (HPLC)

Results & Conclusions

The log10k' for the test substance could not be calculated at either test concentration (5.0mg/L and 10mg/L) because the peak eluted before the sodium nitrate peak, therefore, it is outside the range and accordingly the estimated log10Koc for the test material is reported as <1.5.
