4-Chlorobutyryl chloride was tested in the Ames reverse mutation assay (GLP compliant study according to OECD guidelines 471) using Salmonella typhimurium strains TA98, TA100, TA1535, TA1537 and TA1538 at 20 - 5000 µg/plate (standard plate test) with and without metabolic activation (BASF, 1981). The substance was cytotoxic from 750 µg/plate onward in the standard plate test. Under the conditions tested, 4 -chlorobutyryl chloride was not mutagenic in any of the S. typhimurium strains.

4-Chlorobutyryl chloride was negative for the induction of chromosome aberrations in human lymphocytes (in vitro) in an OECD TG 473 study (BASF, 1992). Positive controls (cyclophosphamide and Mitomycin C) responded as expected.


Short description of key information:
4-Chlorobutyryl chloride did not cause gene mutations in Salmonella typhimurium (Ames test) and in an in vitro cytogenicity test in human lymphocytes.

According to EU Directive 67/548/EEC and EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008 classification is not warranted.
