Optical Brightener 380 is a yellow organic powder. It is characterized by an appreciable molecular weight and carries ionic charges.

There are no empirical bioaccumulation data available on Optical Brightener 380, nevertheless the log Kow resulted to be negative, showing a low affinity with the organic fraction.

Optical Brightener 380 is not toxic/harmful for short-term exposures and it is not skin/eye irritating. It is not able to cause skin sensitization. No genotoxic activity has been observed in specific experiments performed. The subacute repeated dose toxicity study and the screening on reproductive toxicity did not evidence any reason of concern.

There are no specific studies or specific literature data on toxicokinetics and metabolism, thus available information about the substance has been taken into account, avoiding further tests.

Because of the physical state and the trade forms of the substance under registration, inhalation is not an appropriate route of exposure.

Based on physicochemical properties of the substance dermal absorption is expected to be low.  

During the acute oral toxicity experiments run with target substance and Similar Substance 01, the treatment did not produce changes detected in health condition. During the subacute oral tests, treatment with the Similar Substance 01 produced changes in the kidneys and in liver; in particular, macroscopically pale discolouration of the kidneys, increased kidney weights and microscopically renal tubular necrosis and renal inclusion bodies were observed, as well as slight adaptive centrilobular hepatocellular hypertrophy, correlating with enlarged livers were seen at necropsy in some males.

Based on the triazinylaminostilbenes core structure of the substance, it is expected to be stable and not easily metabolized. In addition, the sulphonation degree is expected to contribute to increased solubility and to decreased absorption.
