Description of key information

Propionyl chloride was found to be corrosive to the rabbit skin under occlusive conditions. A single application of undiluted propionyl chloride to the rabbit eye caused cauterization.

Propionyl chloride was applied to the skin of Vienna White rabbits for 1, 5 and 15 min (using 2 animals at all exposure durations) under occlusive conditions and observed for 8 days in a study comparable to OECD guideline 404 (BASF AG, 1969). After 24 hours and 8 days, (severe) necrosis was observed after 1, 5 and 15 min of exposure and erythema (score 2 and 3) was observed at the edges (both animals). The study was not performed for the 20 hours exposure duration, due to the severe effects observed after 1, 5 and 15 minutes exposure. Propionyl chloride was judged corrosive to the skin.

 

Eye irritation

Propionyl chloride was applied to conjunctival sac of the eyes of 2 Vienna White rabbits in a study comparable to OECD guideline 405 (BASF AG, 1969). Animals were observed after 1 and 24 hours and up to 8 days after the treatment. The application caused cauterization of the eyes. Scoring for chemosis, iris, conjunctivae and cornea was not possible. Propionyl chloride was judged corrosive to the eyes.

Justification for classification or non-classification

Based on the available data, propionyl chloride needs to be classified as corrosive to the skin and eyes according to Directive 67/548/EEC (C, R34) and according to the EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008 Category 1B, H314).
