Details on inoculum:
Activated sludge was collected from the exit of the aeration tank, or near the exit from the tank, of a well-operated wastewater treatment plant receiving predominantly domestic sewage.
The dry mass of a known volume of the sludge was determined and the sludge concentrated by removing liquor or diluted further in chlorine-free tap water to obtain the required sludge solids concentration of 3 g.L-1. The activated sludge was continuously aerated (e.g. 2 L/minute) at the test temperature and used on day of collection.
