In an in vitro investigation of cytotoxicity and modulation by hepatic peroxisomes, glycolic acid produced a significant release of LDH from hepatocytes of naïve animals. Administration of clofibric acid increased the release of LDH.

Various investigations into glycolic acid biochemistry give indicators to the mode of action and the underlying chemistry to the effects of glycolic acid: The primary target organ identified is the kidney. Reduced glomerular filtration results from renal damage arising from calcium oxalate monohydrate crystalluria. Histologically this appears as oxalate crystal nephropathy and is apparent in clinical chemistry parameter disturbances in repeated dose studies.
