Study was performed as a limit test at a concentration of 100 mg/L. Sampling was done at 0 hours, 24 hours and after 48 hours. Analysis was done by UPLC-MS.

Daphnia magna was introduced from Nanjing Institute of Environmental Science, MEP in 2018. Culture medium was Elendt M7. All organisms used for a test originated from cultures established from the same healthy stock of Daphnia. The animals were maintained under culture conditions (light, temperature and medium) similar to those used in the test. Animals used for the test were not older than 24 hours.

4 replicates with 5 daphnids in 50 ml of test solution were used in the main test, open test vessels were used and no aeration was provided. The control also consists of 4 replicates with 5 daphnids each.

Number of immobilised daphnids was determined after 24 hours and 48 hours.

Executive summary:

The toxicity of the test item to Daphnia magna was investigated in a limit test with a concentration of 100 mg/l according to Chinese chemical law guidelines and OECD 202. The concentration of the test substance was stable over the test duration of 48 hours. The test fulfills the validity criteria:

  • Number of immobilised daphnids in the control was <10%
  • dissolved oxygen concentration was more than 3 mg/l in control and test vessels
  • the pH values did not differ by more than 1.5 units at test start and end 

Under the test conditions the 48 h EC50 of the test substance was >100 mg/L.
