Assessment of results:
After incubation the screen plates were read using a Titertek Multiscan MCC 340 plate reader and the colour development of the assay medium in each well measured at an absorbance of 540 nm, To correct for turbidity the screen plates were read at a second absorbance level of 620 nm.
Determination of the presence of estrogenic activity was also made by visual assessment of the screen plates and was depicted graphically by plotting corrected absorbance values at 540 nm against test concnetrations (g/L) for each test substance and reference substance.

Applicant's summary and conclusion
- Conclusions:
- D-8 was tested for estrogenic activity in a screening test with a yeast strain containing the human estrogen receptor gene.
The results showed that D-8, tested at top concentrations of 100 mg/L did not demonstrate any evidence of estrogenic activity when screened against the human estrogen acceptor gene contained by the test organism Saccharomyces cerevisiae hER. - Executive summary:
D-8 was tested for estrogenic activity in a screening test with a yeast strain - Saccharomyces cerevisiae hER - containing the human estrogen receptor gene. Visual assessment and photometric measurement of the D-8 screen plates indicated that none of the wells containing D-8 showed any evidence of the conversion of the chromogenic substrate from yellow to red. High levels of estrogenic activity were demonstrated by the positive reference substances 17 ß-estradiol and Bisphenol A. It was concluded that D-8, tested at a top concentration of 100 mg/L, did not demonstrate any evidence of estrogenic activity when screened against the human estrogen receptor gene contained by the test organism Saccharomyces cerevisiae hER.
