- Test concentrations with justification for top dose:
- 0, 2.44, 4.88, 9.77, 19.5, 39.1, 78.1 µg/plate
The highest dose levels were set at 78.1 µg/plate as cytotoxicity to the bacteria was observed. - Vehicle / solvent:
- DMSO
Controls
- Untreated negative controls:
- no
- Negative solvent / vehicle controls:
- yes
- True negative controls:
- no
- Positive controls:
- yes
- Positive control substance:
- 9-aminoacridine
- sodium azide
- other: 2-Aminoanthracene, 2-(2-Furyl)-3-(5-nitro-2-furyl)acrylamide
- Details on test system and experimental conditions:
- The test solution was dispensed into a tube followed by either S9 mix (in the presence of metabolic activation) or Na-phosphate buffer (in the absence of metabolic activation), and each bacterial culture. The content were distributed onto the surface of minimal glucose agar plates. A dose determination test was carried out in a single plate and mutagenicity tests were carried out in triplicates for each dose.
- Rationale for test conditions:
- Guideline study
- Evaluation criteria:
- When the test substance induces a dose-dependent increase in the number of revertant colonies to at least twice as many as that of the negative control and the increase is reproductible, the test substance is judged positive in this test system.
- Statistics:
- none used.

Applicant's summary and conclusion
- Conclusions:
- Throughout the test, the test substance did not induce any increases in the number of revertant colonies to at least twice as many as that of the negative control for any of the bacterial strains at any dose levels either with or without metabolic activation; therefore, the test substance was concluded to be non-mutagenic uner the conditions of this test.
