- RANGE FINDING TESTS:
In Vorversuchen wurde die max. nicht reizende bzw. reizende Konzentration für die intradermale bzw. epidermale Applikation in der Induktionsphase sowie eine nicht reizende Konzentration für die epidermale Applikation in der Provokationsphase an jeweils 2 - 3 Meerschweinchen bestimmt.
MAIN STUDY
A. Intradermale Applikation
Vor der Behandlung der Tiere mit der Prüfsubstanz (Prüfsubstanzgruppe) bzw. mit dem Vehikel (Kontrollgruppe) wurde eine Fläche von ca. 6 x 8 cm der Schulterregion geschoren. Auf einer Flache von 4 x 6 em wurden in 2 parallel verlaufenden Reihen je 3 intradermale Injektionen verteilt. Dabei wurde mit dem ersten Injektionspaar Freunds Complete Adjuvant/Kochsalzlösung (1:1), mit dem zweiten Prüfsubstanzlösung (Kontrolltiere Vehikel) und mit dem dritten Prüfsubstanzlösung und Freunds Complete Adjuvant (1:1) (Kontrolle Vehikel/FCA) verabreicht.

B. Epidermale Applikation
Am 8. Versuchstag wurde die Schulterregion wieder geschoren und das Hautareal mit den Injektionsstellen mit einem Prüfsubstanz- .bzw. Vehikel-Patch okklusiv über 48 Stunden exponiert.Der Patch wurde durch eine Elastikbinde (Acrylastic, P. Beiersdorf und Co. AG., D-2000 Hamburg) fixiert.

C. Provokation
Am 22. Versuchstag wurden die linken und rechten Flanken aller Versuchstiere geschoren (ca. 5 x 5 cm). Allen Tieren der Prüfsubstanz- wie der
Kontrollgruppe wurde auf die linke Flanke die Prüfsubstanz in einer subirritativen Konzentration (höchste nicht reizende Konzentration im Vorversuch) und auf die rechte Flanke das Vehikel aufgebracht und für 24 Stunden jeweils mit einem Okklusiv-Patch (Leukotest®) abgedeckt.

- Nicotinsäure food grade wurde zur Ermittlung seiner sensibilisierenden Eigenschaften an der Haut des Meerschweinchens geprüft. Das Prüfprodukt lag als weisses Pulver vor. Für die Induktionsphase wurde die Prüfsubstanz für die intradermale Applikation mit physiologischer Kochsalzlosung (0.9 % NaCl) auf einen Gehalt von 0.1 % verdünnt und für die epidermale Applikation mit wenig Wasser zu einer Paste verrührt. Für die epidermale Auslösung wurde die Prüfsubstanz mit physiologischer Kochsalzlosung auf 25 % verdünnt.
Nach der epidermalen Auslosung zeigten sich 24 nach Abnahme des Okklusiv-Patches weder bei den Tieren der Behandlungs- noch bei den Tiere der Kontrollgruppe Hautreaktionen.
Nicotinsaure food grade besitzt aufgrund dieser Prüfung keine sensibilisierenden Eigenschaften an der Haut von Meerschweinchen (s. Kennzeichnungsleitfaden der EG (2, 9).
Systemisch-toxische Eigenschaften traten nicht auf. Allgemeinbefinden und Körpergewichtsentwicklung der Versuchstiere waren unbeeinflusst. - Executive summary:
A study was carried out according to EU Method B.6 and OECD Guideline 406 (Skin Sensitisation), assess skin sensitizing properties of the test item in a guinea pig maximization test. Following epicuteaneus challenge no sensitisation effects were observed 24 and 48 hours after removal of the exposure patch, either in the treatment or in the control group. Therefore, the test item is considered not sensitizing to skin.
