Toxicity to soil macroorganisms except arthropods

Description of key information

The chronic toxicity of the test substance to earthworms (Eisenia fetida) was studied according to OECD TG 222 and in compliance with GLP criteria. Method: Nine treatment groups were tested including control, and test item treatment with nominal concentrations corresponding to3.27, 5.88, 10.6, 19.1, 34.3, 61.7, 111 and 200 mg test item/kg soil dry weight. The test was performed using 4 replicates for the test item treatments and 8 replicates for the control, with 10 earthworms per replicate. The earthworms were exposed in artificial soil for 8 weeks. The adult earthworms (about 10 months old with clitellum; weighted 319 – 599 mg) were acclimated in an artificial soil for 24 hours before exposure. One day after application, 5 g dry ground cattle manure was scattered on the soil surface of each test vessel, and sprinkled with 5 g deionised water. The feeding interval was weekly during the first four weeks of the study. After four weeks, the adult worms were removed. The number of surviving worms and the bodyweight of worms per vessel was determined. The juvenile worms remained in the test vessels for a further 4 weeks. When the test was terminated, the number of survival, behavioural and pathological symptoms of the juvenile worms were recorded. The test conditions were maintained at 18 - 22 ˚C; 16 - hour light and 8 - hour darkness (light intensity: 400 - 600 lux), and pH 5.7 – 6.1. Results: No behavioural abnormalities were observed. Mortality was seen at > 111 mg test item/kg soil. The NOEC for weight changes was determined to be ≥200 mg test item/kg soil. The biologically relevant NOEC for reproduction is considered to be the concentration of 19.1 mg test item/kg soil. The EC10 for reproduction was determined to be 16.1 mg test item/kg soil (95% confidence limits of 0.41 to 32.7 mg test item/kg soil).
