Description of key information

Freshwater aquatic toxicity studies with Glycerol 1,3 dimethacrylate conducted in fish. Aquatic toxicity in invertebrates and alga were calculated by OECD Toolbox based on valid own company experimental data of methacrylates with an unspecific mechanism.

The measured log Kow is 2.05. QSAR predictions with OECD toolbox version 3.3 are performed KOWWIN 1.68 which calculates a log Kow of 1.16.

The QSAR prediction of acute fish toxicity was performed to demonstrate on the one hand that the predicted LC50 of 133 mg/l is higher than the measured but primarily that the experimental LC50 fish of 43.2 mg/l correlates with the measured log Kow. Based on this correlation in a further step QSAR predictions for daphnia magna and algae were performed (see 6.1.3 and 6.1.5) and the corrected EC50 values were graphically read for the measured logKow.

 

Short term toxicity to fish:

The acute lethal toxicity (LC50) of GLYCEROL-1 ,3-DIMETHACRYLATE to Poecilia reticulata (Guppy) was investigated under semi-static exposure conditions over a period of 96 h acc. OECD 203

Result: LC50 (96h): 43.2 mg/1 (based on nominal concentrations).

Short-term toxicity to aquatic invertebrates

EC50 Daphnia magna (48h) of Glycerol 1,3 dimethacrylate (199 mg/L) is predicted from category members using trend analysis based on 8 values within the range 4.56 - 380 mg/L from 8 category members. Category members are single substances or mixtures and are selected based on the profile of the target chemical. Only substances with vaild experimental data are listed in the category.

The target chemical FALLS within applicability domain of the prediction.

 

The predicted value of 199 mg/l is based on Log Kow 1.16 which is calculated by KowWin.

Glycerol 1,3 dimethacrylate is an ether methacrylate and log Kow of ethers are known to be difficult to calculate. The measured log Kow is 2.05. The QSAR prediction was used to create the graphic which shows the correlation of the acute toxicity to daphnia magna of the different methacrylates and log Kow. (see also 4.7 Partition coefficient).

 

The correlation of EC50 is consistent with increasing log Kow and therefore the diagram was used to read the EC50 Daphnia magna of Glycerol 1,3 dimethacrylate with log Kow 2,05.

An EC50, log (1/mol/l) of 3.05 was read from the y-axis of the graphic which recalculated corresponds to an EC50 Daphnia magna (48h) of 80 mg/l.

Toxicity to aquatic algae

 

Toxicity of the Glycerol 1,3 dimethacrylate (158 mg/L) is predicted from category members using trend analysis based on 13 values within the range 2.25 - 836 mg/L from 13 category members. Category members are single chemicals or mixtures and are selected based on the profile of the target chemical. Only chemicals having experimental data are listed in the category.

The target chemical FALLS within applicability domain of the prediction. The data used for calculating the current prediction is taken from 14 experimental values

 

The predicted value of 158 mg/l is based on Log Kow 1.16 which is calculated by KowWin.

2-hydroxy-1,3-propanediyl bismethacrylate is an ether methacrylate and log Kow of ethers are known to be difficult to calculate. The measured log Kow is 2.05. The QSAR prediction was used to create the diagram which shows the correlation of the acute toxicity to alga of the different methacrylates and log Kow. The correlation of EC50 was consistent with increasing log Kow therefore the graphic was used to find the EC50 Alga of  2-hydroxy-1,3-propanediyl bismethacrylate with log Kow 2.05.

An EC50, log (1/mol/l) of 3.55 was read from the y-axis of the diagram  which  recalculated corresponds to an EC50 of 64 mg/l.

 

Chronic toxicity alga:

The predicted value of 68,9 mg/l is based on Log Kow 1,16 which is calculated by KowWin. 2-hydroxy-1,3-propanediyl bismethacrylate is an ether methacrylate and log Kow of ethers are known to be difficult to calculate. The measured log Kow is 2.05. The QSAR prediction was used to create the diagram which shows the correlation of the chronic toxicity to alga of the different methacrylates and log Kow. The correlation of NOEC  was consistent with increasing log Kow therefore the diagram was used to read NOEC algae of  2-hydroxy-1,3-propanediyl bismethacrylate with log Kow 2.05. A NOEC, log (1/mol/l) of 4.05 was read from the y-axis of the diagram which  recalculated corresponds to a NOEC of 20 mg/l.
