The concentration verification of the test solution was carried out at 0 hour and 48 hours for active substance 1-Benzyl-5-Phenylbarbituric acid (BPBA) after initiation of exposure, at the concentration of 50 mg/L (Maximum Soluble Concentration).
At 0 hour percentage difference observed from the initial concentration for 1-Benzyl-5-Phenylbarbituric acid (BPBA), at the concentration of 50 mg/L was 7.98.
At 48 hours percentage difference observed from the initial concentration for 1-Benzyl-5-Phenylbarbituric acid (BPBA), at the concentration of 50 mg/L was -1.36. - Details on test conditions:
- EXPERIMENTAL PROCEDURE
The test was conducted in beakers of capacity 100 ml. The gravid females were separated from the culture with the help of a pasteur pipette in glass bottles filled with reconstituted water. Newborn daphnids / neonates were sampled at random with a pasteur pipette and carefully transferred into the test containers. Twenty daphnids were grouped into four replicates (5 Daphnids per replicate). One concurrent blank control group was run in addition to the test series.
Reference item control using Potassium dichromate was maintained with three doses 0.40, 0.63 and 1.00 mg/L with log interval of 0.2.
OBSERVATIONS
PHYSICAL PARAMETERS
Measurement of dissolved oxygen, pH and temperature of the test water was measured at the beginning and at the end of the test in the control and in the highest test and reference item concentration group during the exposure period of 48 hours. Hardness of reconstituted water was also measured.
IMMOBILISATION
The daphnids were observed for immobility at 24 and 48 hours of exposure. Those daphnids which were unable to swim within 15 seconds after gentle agitation of the test container were considered as immobile.
EC50 CALCULATION
The percentage immobility at 24 and 48 hours was calculated by Maximum Likelihood Method for EC50 calculation described in ‘Probit Analysis’ by D. J. Finney (1971). - Reference substance (positive control):

Acute Immobilisation Test of 1-Benzyl-5-Phenylbarbituric acid (BPBA) in Daphnia magna was performed in compliance with the Organization for Economic Co-operation and Development (OECD) Guideline for testing of chemicals, No. 202, 'Daphnia Sp., Acute Immobilisation Test' , Section 2, adopted by the council on 13 April 2004.
Test item was suspended in reconstituted water and was tested for acute toxic effects on the swimming capabilities of Daphnia magna employing a static test system. In the main study, four replicates per treatment group with 5 daphnids per replicate were exposed to the 1-Benzyl-5-Phenylbarbituric acid (BPBA) at concentration of 50 mg/L (Maximum Soluble Concentration). Observations were made for immobilisation at 24 and 48 hours of treatment. A concurrent blank control was also run simultaneously. Potassium dichromate was used as a reference item and tested at the concentrations of 0.40, 0.63 and 1.00 mg/L.
No immobilisation was observed within the 24 and 48 hours at the concentration of 50 mg/L (Maximum Soluble Concentration). No immobilisation was observed in blank control group.
The percentage of daphnids those were immobilized in the reference item group at 24 hours were 15, 50 and 80 and at 48 hours were 20, 55 and 100 at the concentrations of 0.40, 0.63 and 1.00 mg/L respectively.
The acute immobilisation EC50 value of 1-Benzyl-5-Phenylbarbituric acid (BPBA) in Daphnia magna at 24 and 48 hours exposure period is estimated to be greater than 50 mg/L (Maximum Soluble Concentration).
