In vitro studies ( Salmonella typhimurium mutagenicity-tests):
Two studies regarding mutagenicity in bacterial test systems (Ames test) of the test item Isophorone nitrile were conducted. (TNO, 1990; Hüls AG, 1985). Altrough both studies have the same reliability and leading to the same conclusions, the newer study (TNO, 1990) was choosen to be the key study because this study was performed under GLP conditions. According to the outcome of both studies it has to be concluded that the test item Isophorone nitrile is not mutagenic under the conditions of these studies.
In vivo study (mouse micronucleus test):
An in vivo mouse micronucleus test was conducted with the test item Isophorone nitrile (Asta, 1992). Under the conditions of this study the test item Isophorone nitrile was not mutagenic.
Short description of key information:
The test item 3-cyano-3,5,5-trimethylcyclohexanone (Isophorone nitrile) was not mutagenic under conditions of Salmonella typhimurium mutagenicity-tests (Ames-Tests; TNO, 1990 and Hüls AG, 1985) and it was not mutagenic under conditions of the conducted in vivo mouse mironucleus test (Asta, 1992).
Endpoint Conclusion: No adverse effect observed (negative)
Justification for classification or non-classification
Regarding mutagenic properties the substance 3-cyano-3,5,5-trimethylcyclohexanone (Isophorone nitrile) is not classified according to the criteria of EC Directive 67/548/EEC.
