To investigate the toxicity of the test substance to aquatic algae a GLP guideline study according OECD 201 including analytical monitoring has been conducted (RCC Umweltchemie GmbH & Co. KG (1995c). Since the test substance is coloured the test method was modified to differentiate between a reduced growth of algae due to real toxic effects of the test substance on the algal cells or due to an indirect effect, a reduced algal growth by light absorption in coloured test media. Two different test set ups were started. In the part A the algae were directly exposed to the test item in 5 test concentrations (prepared with the emulsifier Tween 80) to determine possible inhibitory effects of the test substance. In the second experiment B, glass dishes including the same test concentrations as in part A covered the Erlenmeyer flask where the algae grew in media including the emulsifier Tween 80 but without test substance. Therefore, effects due to the filter effects of the coloured test media could be measured.

After 72 h of exposure the same or even a higher inhibition of the growth of Scenedesmus subspicatus was observed when the algae grew in test water without test substance, but under reduced light intensities by the filter effect of the coloured test media as in the second parallel experimental part, where the algae grew in the test media with suspended test substance. In conclusion, this modified algal test has clearly demonstrated that the observed growth inhibition effect of the test substance on Scenedesmus subspicatus was caused only due to the indirect effect, the light absorption in the coloured test media. Thus, a real toxic effect of the test substance on the algal cells can be excluded up to at least the highest tested concentration of 100 mg test substance/1, and thus far above the water solubility limit of the test substance.
