Results and discussion
- Preliminary study:
- All animals were fasted overnight prior to and for one hour after dosing. Water was freely available at all times. The test article was administered as a single oral dose by gavage to five groups of two male and two female rats at dose levels of 5000, 1600, 500, 160 and 50 mg/kg. The range of intital weights was 180-203 g for males and 189-204 g for females. All animals were observed for 3 days for mortality. No deaths were observed in males dosed with 50 or 160 mg/kg. All males dosed with 500, 1600 or 5000 mg/kg died within 30 min of dosing. No females died at 50 mg/kg; one female died at 160 mg/kg and all females died at higher doses within one hour.

all animals in the 300 and 500 mg/kg dose groups
All deaths occurred within 24 hours post-treatment. - Clinical signs:
- other: lethargy, salivation, ataxia, lacrimation, soft feces and squinted eyes. No significant clinical observations were seen in animals living past day one (one female exhibited extended salivation, rhinorrhea and wheezing).
- Gross pathology:
- of animals found dead:
LUNGS: dark areas (8/27), discoloration (13/27), mottling (10/27), congestion (5/27)
SPLEEN: dark areas (3/27)
STOMACH: distention with compound colored fluid (14/27), distention with fluid (10/27), distention with gas (15/27)
INTESTINES: distention with gas (2/27) and fluid (14/27)
In animals sacrificed on Day 14:
discoloration of, dark red areas and white foci on the lungs, a dark red and slightly enlarged uterus in a female in the 64.5 mg/kg dose group adn a cyst and fluid in the uterus of a female in the 107.5 mg/kg dose group. - Other findings:
- none

Applicant's summary and conclusion
- Interpretation of results:
- toxic
- Remarks:
- Migrated information Criteria used for interpretation of results: US CPSC / US OSHA
- Conclusions:
- oral LD50 rats =174 mg/kg
- Executive summary:
Crotonaldehyde was testing in sprague dawley male and females rats for acute oral toxicity by gavage. The test stubstance was administered in corn oil. Body weights and pathology were conducted on all animals as well as clinical signs observations. The result of the study was that the oral LD50 in rats (male and female combined) is 174 mg/kg.
