It is concluded that neither Ethyldehydrolinalool per se, nor
any of the metabolites formed under the described experimental conditions is
mutagenic in the Ames test.
Executive summary:

Ethyldehydrolinalool was evaluated for mutagenic activity in the Ames test. A standard plate incorporation and a preincubation modification assay in absence and in presence of an exogenous metabolic activation system (S9) were

performed. Five Salmonella typhimurium tester strains (TA1535, TA97, TA98, TA100, and TA102) were employed. The activity of the S9-mix and the responsiveness of the tester strains were verified by including appropriate controls into each experiment.

Ethyldehydrolinalool was dissolved in dimethylsulfoxide (DMSO). Strong toxic effects were observed in a preliminary toxicity experiment starting at 1’666 ug/plate (no growth). Therefore concentrations ranging from 10 to 1’000 ug/plate were

evaluated in the main experiments. No precipitation of the compound was observable

up to the highest test concentration of 1’000 ug/plate using the standard procedure,

whereas a slighty turbidity at the top dose was observed with the preincubation modification assay. Strain dependent toxic effects were observed (reduction in the number of-revertants and/or reduction in the background growth). Toxicity was more

pronounced using the preincubation method, known to be more sensitive for several class-of-compounds.

No increase of the number of mutant colonies was apparent in all five investigated tester strains.
