Details on test organisms:
TEST ORGANISM
- Strain: 61.81 SAG (identical strains: CCAP 278/4; UTEX 1648; ATCC 22662)
- Source: The algae were supplied by the SAG: Collection of Algal Cultures, Inst. Plant Physiology, University of Göttingen, Untere Karspüle 2, D-37073 Göttingen, Germany
- Stage: exponential growth
- Method of cultivation: The stock cultures are small algal cultures that are planted on agar. These are transferred to fresh medium at least once every two months under standardised conditions according to the test guidelines. The pre-culture was prepared with OECD Medium, incubated under the conditions of the test and used when still exponentially growing, normally after an incubation period of 2-4 days. (The pre-culture was incubated for four days at this test.)

- Unusual cell shape: no
- Colour differences: no
- Flocculation: no
- Adherence to test vessels: no
- Aggregation of algal cells: no
- Any stimulation of growth found in any treatment: no
- Any observations that might cause a difference between measured and nominal values:
- Effect concentrations exceeding solubility of substance in test medium: no
Results with reference substance (positive control):
72h ErC50: 0.62 mg/L, (95 % confidence limits: 0.59 – 0.65 mg/L); 72h EyC50: 0.33 mg/L, (95 % confidence limits: 0.32 – 0.34 mg/L)
Reported statistics and error estimates:
The ECx values and their 95 % confidence limits for growth rates and yield were calculated by Probit-analysis using linear max. likelihood regression. For the determination of the NOEC/LOEC values, Step-down Jonckheere-Terpstra test (alpha = 0.05, 2-sided) was performed with growth rates and yield. Pretesting on normal distribution was performed by Shapiro Wilk’s test and on variance homogeneity by Levene’s test.

VALIDITY OF TEST

Cell density in the control cultures increased by a factor of more than 16 (by a factor of 104.0) within 72 hours. This corresponds to a specific growth rate of 1.54 day-1.

The mean coefficient of variation (CV) for section-by-section specific growth rates (days 0-1, 1-2 and 2-3) in the control cultures did not exceed 35 %.

The mean CV for section-by-section specific growth rates: 20.4 %. The coefficient of variation (CV) of average specific growth rates during the whole test period in replicate control cultures did not exceed 7 % in test.

The CV for average specific growth rate (day 0-3): 4.3 %

All validity criteria were met, therefore the study can be considered as valid.

Validity criteria fulfilled:
yes
Conclusions:
In a Growth Inhibition Test with Raphidocelis subcapitata according to OECD 201 the 72-h EC10 and EC50 of the test item for growth rate were determined to be 6.75 and 15.55 mg/L, respectively. The 72-h EC10 and EC50 for yield were determined to be 1.89 and 7.80 mg/L, respectively. The 72-h NOEC for growth rate and yield was determined to be 4.84 mg/L. All biological results are based on geometric mean measured concentrations.
Executive summary:

To determine the effect of the test item on the growth of a unicellular green algal species a Growth Inhibition Test according to OECD 201 and EU Method C.3 was carried out in compliance with the GLP principles. Exponentially growing cultures of Raphidocelis subcapitata (previously known as Pseudokirchneriella subcapitata) were exposed for a duration of 72 hours to nominal test item concentrations of 0.30, 0.95, 3.1, 9.8, 31.3 and 100 mg/L. For determination of the test item concentrations, samples were taken from each concentration level and from the control at the start and at the end of the test. Using a HPLC-UV the geometric mean test item concentrations of the measured start and end concentrations were calculated to be 0.19, 0.49, 0.87, 4.84, 25.22 and 88.93 mg/L. Biological results and endpoints are thus based on the measured concentrations. The test design included three replicates at each test concentration and six replicates for the untreated control. The alga cell concentration was approximately 104 cells/mL at the start of the test in all of the test cultures. Glass flasks with total capacity of 250 mL were used as test vessels. The algal cell concentration was determined by manual cell counting by microscope in each testing flask during the 72-hour test, in 24-hour intervals. The results of the statistical evaluation showed that the 0-72 h average specific growth rate and yield was not statistically significantly different from the control in the exposure concentrations of 0.19, 0.49, 0.87 and 4.84 mg/L and significantly different in the exposure concentrations of 25.22 and 88.93 mg/L. The 72-h EC10 and EC50 of the test item for growth rate were determined to be 6.75 and 15.55 mg/L, respectively. The 72-h EC10 and EC50 for yield were determined to be 1.89 and 7.80 mg/L, respectively.  The 72-h NOEC for growth rate and yield was determined to be 4.84 mg/L. All validity criteria of the guidelines were met; the study is hence considered valid.
