Experimental studies investigating acute toxic effects of the target substance Sorbitan tridocosanoate on fish are not available. Thus, in accordance with Regulation (EC) No 1907/2006 Annex XI, 1.5 a read across to the structurally related source substances Sorbitan laurate (CAS 1338-39-2) and Sorbitan stearate (CAS No.1338-41-6) was conducted to fulfil the data requirements according to Annex VIII. The most suitable (highest degree of structural similarity, nearest physico-chemical properties) read-across substance was used for the assessment. The read-across approach is justified in detail within the analogue justification in IUCLID Section 13.
The study on the source substance Sorbitan laurate (CAS 1338-39-2) was performed as a limit test, according to OECD Guideline 203 and GLP. Fish (Danio rerio) were exposed to a nominal test concentration of 100 mg/L, prepared as water accommodated fraction (WAF). No mortalities were recorded during the 96-hour test period. The determined LC50 (96 h) is > 100 mg/L.
The available study on the source substance Sorbitan stearate (CAS 1338-41-6) was conducted as a limit test, according to the Circular on Test Methods of New Chemical Substances (Japan). A semi-static system was set up. The fish (Oryzias latipes) were exposed to a nominal test concentration of 1000 mg/L, prepared as water accommodated fraction (WAF). An analytical dose verification was performed using LC/MS measurements. The measured concentration was 13.7 mg/L (TWA), which is far above the determined water solubility of < 0.525 mg/L at 20 °C. Due to the fact that the substance is an UVCB and the preparation of a WAF it was considered to be more appropriate to use the nominal loading rate for the assessment. No mortalities were observed during the 96-hour test period. The recorded LL50 is > 1000 mg/L based on the nominal loading rate
