As the solubility of test item in the buffer waters was more than > 10-2 g/L, "Shake Flask Method" was adopted to determine its solubility in buffer waters.

Conclusions:
The solubility of the test substance in three different buffers (pH=5, 7 and 9) is assumed to be below the detection limit of analytical method, i.e. 0.23 mg/L.


Executive summary:

Determination of water solubility of the test substance was performed in accordance with EEC method A.6 and OECD 105. About five times higher quantity of test item determined in the preliminary test were weighed and transferred into three separate glass bottles of 125 mL capacity containing each 50 mL of buffer solution (pH=5). The same was repeated for buffer solution of pH=7 and pH=9. All the nine bottles were tightly stoppered and agitated at 30ºC using a thermostatically controlled orbital shaker. After 1 day, 2 days and 3 days, bottles were removed and kept for re-equilibration at 20ºC for 24 hours. The bottles were shaken occasionally during this period. Then the content of the bottles were separated using syringe having removable needle. The pH of solution was measured using a calibrated pH meter. The active ingredient concentration analysis was carried out using a validated analytical method. The solubility of the test substance in all three buffers was <LOD (0.23 mg/L). There was no chromatographic peak of any of the two components (viz. laevo alpha pinene and camphene) observed in shake flask analysis due to their negligible (very low) solubility in water. Thus, the solubility of the test substance in three different buffers (pH=5, 7 and 9) is assumed to be below the detection limit of analytical method, i.e. 0.23 mg/L.
