Naphtol AS PH did not increase the number of revertant colonies in all tested strains of bacteria.

The number of micronuclei in normochromatic as well as polychromatic erythrocytes of the bone marrow of orally treated mice was unchanged.

The positive control substances provoked the expected effects indicating the sensitivity of the test systems.


Short description of key information:
Potential mutagenic effects of Naphtol AS PH were examined using the Ames bacterial mutagenicity test in Salmeonella typhimurium (Strains TA 98, TA 100, TA 1535, TA 1537, TA 1538) and Escherichia coli (strain: WP2uvrA) as well as the mouse micronucleus test in vivo.
None of the bacterial strains showed a relevant increase of the number of revertant colonies after treatment in the presence as well as the absence of a metabolic activating system (rat liver S9 mix). The positive as well as the negative control cultures exhibited the expected results.
Oral application (gavage) to male and female mice

Endpoint Conclusion: No adverse effect observed (negative)

Justification for classification or non-classification

No mutagenic effects were detected in vitro in bacteria as well as in vivo in mouse bone marrow cells.
