In an OECD guideline 404 study (BASF AG, 1990) three rabbits were treated with the undiluted test substance for 4 h using semiocclusive conditions. The animals were observed for 15 days and skin changes were recorded. The test substance caused well-defined edema and moderate erythema 24, 48 and 72 hours after application, which were fully reversible within 15 days. In all animals scaling was observed at the end of the observation period (first noticed on day 8). Based on the results, the test substance was identified as an irritant.

This result is supported by the outcome of an older skin irritation test (BASF AG, 1977) where the test substance – applied occlusively on the skin of 2 rabbits for 1, 5, 15 min or 20 hrs – induced erythema and edema of grade 3 at the 24 h reading. At the next reading (after 8 days) these skin reactions had disappeared but desquamation was present in both animals.

 

Eye irritation

In an OECD guideline 405 study (BASF AG, 1990) the right eyes of three rabbits were treated with 0.1 mL of the test substance. The untreated left eyes served as control. The animals were observed for 16 days and eye changes were recorded at 24, 48, 72 hrs and at 8 and 16 days. The application of the test substance caused distinct reddening and edema formation , well-defined (conjunctivae score / chemosis score) and moderate (iris score/cornea score) eye reactions were observed within 24 - 72 hours after application, which were fully reversible within 16 days (reddening) or within 8 days.

In an earlier study (BASF AG, 1977) 50 µL of the test substance was applied to the eye of two rabbits and eye reactions were recorded 1 h and 24 hrs after test item instillation and at the end of the 8-d observation period. Also in this study the test substance was irritating to the rabbit eye.

Based on the results obtained in the skin and eye irritation tests, 2 -Methylpentanal is classified as follows:

 

R38, irritating to skin (according to Directive 67/548/EEC) and H319, causes serious eye irritation, Cat. 2, and H315, causes skin irritation, Cat. 2 (according to Regulation 1272/2008/EC).
