Toxicokinetics

The toxicokinetics of 3a,4,5,6,7,7a-hexahydro-4,7-methanoinden-6-yl acetate have been investigated on dermal studies. Following oral administration and intra-arterial in fusion, 3a,4,5,6,7,7a-hexahydro-4,7-methanoinden-6-yl acetate is found to be rapidly absorbed and is excreted predominantly in the urine. Based on the study reported it indicates that there is no potential for bioaccumulation.

 

Acute toxicity

 

3a,4,5,6,7,7a-hexahydro-4,7-methanoinden-6-yl acetate is of low acute toxicity by all routes investigated, the acute oral LD50 of the substance was found to be 5400 mg/kg/bw on the mouse (Pergamon Press Inc., Maxwell House); the acute dermal also do not show any major deviation from the oral route and show the LD50 value as > 5000 mg/kg. The acute inhalation toxicity is not available for the target substance and so the same have been considered for the read across substance and the same also do not qualify in toxic range.

 

Irritation / corrosion

In vivo studies of the substance show that the substance is a non - irritating to skin and also do not have irritation effect to eyes, and so the substance is not classified in skin irritation category but have be categorized in class 2 for eye irritation according to current criteria.

 

Sensitisation

No evidence of skin sensitisation was noted in a estimation study for the substance. In two separate maximization tests involving 26 and 21 volunteers, there was no evidence of contact sensitization to 20% (13,800 lg/cm2)or 8% (5520 lg/cm2) tri cyclo decenyl acetate(3a,4,5,6,7,7a-hexahydro-4,7-methanoinden-6-yl acetate), respectively.

3a,4,5,6,7,7a-hexahydro-4,7-methanoinden-6-yl acetateis of non acute toxicity, is slight irritant, is not genotoxic and is not a developmental or reproductive toxin.
