Description of key information

The aquatic toxicity of 3-[[3-(dimethylamino)propyl]amino]propiononitrile was determined in accordance with GLP and OECD test guidelines. The tests included four trophic levels of organisms fish,Daphnia, algae and STP microorganisms.

In accordance with the recommendations of REACh, the acute fish test was conducted according to the threshold approach recommended by ECHA. Using this approach the lowest EC50value from either the Algal Growth Inhibition study or Acute Toxicity toDaphnia magnastudy is set as the threshold concentration and a “Limit test” is conducted at this threshold concentration.

The test substance was found to be stable in aquatic media by LC/MS analysis, all measured test concentrations were maintained with the range 80 – 100 range and therefore endpoints were reported based on nominal concentrations.

The toxicological endpoints were:

Acute fish test (Oncorhynchus mykiss) LC50(96h): >62 mg/L, NOEC: ≥ 62 mg/L

AcuteDaphnia magnatest: EC50(48h): >100 mg/L, NOEC: 100 mg/L

Algal inhibition assay (Pseudokirchneriella subcapitata): ErC50(72h): 62 mg/L, ErC10(72h): 36 mg/L, NOEC (72h): 12.5 mg/L

Activated sludge respiration: EC50(3h): >1000 mg/L

It is concluded that the most sensitive species was the algal species and the endpoints for regulatory use and PNEC derivation will be based on the ErC50(72h): 62 mg/L, ErC10(72h): 36 mg/L.
