- El ensayo in vitro de irritación la piel se utiliza para determinar la irritación de la piel de los productos químicos mediante la medición de la viabilidad celular en una epidermis humana reconstruida (RHE) modelo después de aplicar la sustancia de ensayo.
El modelo RHE consta de queratinocitos epidérmicos origen humano, que han sido cultivados para formar un modelo de múltiples capas de la epidermis humana. Se identifican irritantes químicos por su capacidad para disminuir la viabilidad celular por la medición cuantitativa de la conversión enzimática de la vital MTT colorante (3 - [4,5-dimetil tiazol 2-il] 2,5-difenil-tetrazoliumbromide) a un azul formazán.
El potencial de irritación cutánea de la sustancia de ensayo se evalúa mediante la reducción de la producción de formazano a menos del 50% del control negativo. - GLP compliance:
