Fish were sampled from rivers near textile and dyestuff manufactures known to use certain aromatic amines. To determine the amines fish tissue was digested and extracted with benzene. After separation amines were quantified by gas-liquid chromatography and recoveries determined. The technique employed aimed at detecting less water-soluble aromatic amines which are expected to biomagnify or concentrate to higher levels in fish tissue than are found in the aqueous environment.

Study design

Details on sampling:
Fish near dye or textile manufacturing plants were collected by various stage agencies either by netting or electroshock technuques, and immediately placed on ice or dry ice. Frozen fish were filleted, doubly ground, and thoroughly mixed. Fish were stored frozen if immediate analysis was not possible.

N-phenyl-1-naphthylamine was not detected in the fish collected from the surface water sites close to the dye or textile manufacturing plants.

In control experiments to test the efficiency of the method employed Ocean Perch tissue was spiked with the test substance at levels ranginng from 20 to 100 ppb (edible portion). The average recovery found was 90% (7.1 average deviation from mean; 8.4 standard deviation). By aid of nitrogen-selective gas chromatography 14 ng of N-phenyl-1-naphthylamine was detected from fish tissue spiked at 80 µg//kg.
