Details on test solutions:
The test solutions were prepared by mechanical dispersion, without the use of any solubilising agent. The test item is poorly water soluble material. In order to achieve maximum dissolved concentration, a supersaturated solution was prepared by dispersing/dissolving the test item amount of 0.76 g into 1000 mL test medium (OECD medium; see 5.4) one day before the start of the test. This test item solution was agitated by orbital shaker (~300 rpm) for a period of about 20 hours and then the non-dissolved test material was separated by filtration through a fine (0.22 μm) filter in order to obtain the saturated test solution (i.e. 100 % v/v saturated solution). The further test solutions were freshly prepared from this test item stock solution (TISS) in the testing laboratory just before introduction of algae (start of the experiment) by appropriate diluting of this stock solution.

- Breeding Conditions: The stock cultures are small algal cultures that are planted on agar regularly. These are transferred to fresh medium at least once every two months under standardized conditions according to the test guidelines.
- Pre-culturing: The pre-culture is intended to give an amount of algae suitable for the inoculation of test cultures. The pre-culture was prepared with Algal Mineral Salts Culture Medium, incubated under the conditions of the test and used when still exponentially growing, normally after an incubation period of about three days. (The preculture was incubated for four days at this test.) The algal cultures used in this study did not contain deformed or abnormal cells.

Based on the results of a preliminary tests, the test solutions were prepared from a test item stock solution (TISS) at the concentration equal to the water solubility (water accommodated fraction, i.e. WAF) and the following test item concentrations were used in the main study: TISS/39.0625; TISS/15.625; TISS/6.25; TISS/2.5 and TISS.
The analytically measured test item concentrations deviated more than ± 20 % of the initial test item concentrations over the test period of 72 hours, therefore the biological results are based on the measured concentrations.
The corresponding calculated geometric mean test item concentrations (based on the analytical measurements) were NA1*, NA2*, 0.03, 0.06 and 0.16 mg/L.

Results with reference substance (positive control):
For the evaluation of the reliability of the applied test system and experimental conditions, potassium dichromate is tested at least twice a year. The date of the last study with reference item Potassium dichromate was: September 23 - 26, 2014.

Mean values and standard deviations were calculated for each treatment at the start, 24 and 48 hours after treatment and at the end of the test using Excel for Windows software.
For the determination of the LOEC and NOEC, the calculated mean growth rates and yield at the test concentrations were tested on significant differences to the control values by Bonferroni t-Test (α = 0.05) using TOXSTAT software. The data were checked for normality by Chi-Square Test, for homogeneity of variance by Hartley’s Test.
As the inhibition of growth of the algal cells was lower than 50 % regarding growth rate and yield, there was no hence to perform Probit analysis.
