Several special investigations in animals have been conducted to evaluate the neurotoxicity potential of 2-pyrrolidone.

In one study, 2-pyrrolidone was applied intravenously in a single dose to mice (Hawkins and Sarett, 1957). Control animals received vehicle on the same routes. No antivconvulsive effects were observed against elektro-shock induced convulsions. Interestingly convulsions induced by beta-methyl-beta-ethylglutarimide or pentylenetetrazole were effected and the anticonvulsive ED50 was 150 and 200 mg/kg bw respectively. Another study revealed a tranquilising potential of 2 -pyrrolidone (Siroslawska, 1965). Mice, rats, rabbits, cats or guinea pigs which received intraperitoneally 2 -pyrrolidone exhibited significant slower movements, a reduced ability for coordination and a reduced spontaneous activity in the brain.

The youngest study available dealt with the investigation whether analogons of 4-hydroxy butyric acid and butyrolacton would affect the balance reaction, and revealed that they did not influence the rightening reflex in rats (Markus et al., 1976). Beside that the study revealed a desynchronisation in the EEG after administration of the analogons.

Lightowler et al. investigated 1963 in mice, whether 2-pyrrolidone had anti-strychnine activities (Lightowler et al., 1963). The study revealed that 2-pyrrolidone had anti-strychnine activity at 875 and 1750 mg/kg but not at higher doses.

Justification for classification or non-classification

Classification is not warranted according to the criteria of EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008.
