Description of key information

Short-term and long-term toxicity endpoints are available for a number of freshwater aquatic species. Studies are available for three different trophic levels covered by fish, daphnia and algae. Long term studies are presented for the affect of a Bardap 26 structural analog surrogate, Bardac 22 (DDAC), on zebra fish survival, and daphnia reproduction and survival. A long term study was also carried out to investigate the impact of Bardap 26 on the growth of a unicellular algae (Scendesmus). From the results obtained, daphnia emerge as the most sensitive species with a 21-day NOEC for survival of 0.01 mg/l.

No testing on marine species was carried out as Bardap 26 is used in the formulation of cleaning products, water treatment products and metal treatment products and thus, marine water is not justified.

Short- term toxicity to fish:

A 96-hour acute toxicity study was conducted to determine the ability of N,N-Didecyl-N-methylpoly(oxyethyl)ammonium Propionate to cause acute toxic effects to fish when tested in accordance with EPA OPPTS 850.1075. The test animals used were Bluegill (Lepomis macrochirus), exposed to the test material for 96 hours in a flow-through test system at concentrations of 0, 0.10, 0.20, 0.40, 0.80 and 1.6 mg a.s./l. (nominal concentrations: 0, 0.090, 0.19, 0.38, 0.77 and 1.4 mg/L).

100% mortality was observed at the 0.77 and 1.4 mg mg/L concentration groups and 5% mortality was recorded in the 0.38 mg/L test group. No adverse effects were observed in any test group in the surviving fish. Based on concentration effect relationship observed, under the conditions of this study, the no-observed-effect concentration (NOEC) was found to be 0.19 mg/L.

 

Long-term toxicity to fish:

In a study conducted in accordance with OECD Guideline No. 210, Zebra fish (Brachydanio rerio) were exposed to the test material, Bardac 22 with Didecyldimethylammonium Chloride, for 34 days at concentrations of 0, 1.0, 3.2, 10, 32, 100 and 320 mg/L. 4 replicates of 20 eggs/larvae were dosed per concentration.  Hatching, mortality, morphological and behavioural development, body weight and body length were the test parameters examined. The test substance had no effect on hatching. All fish died at a test substance concentration of 320 µg/L. At 100 µg/l fish were observed swimming on the surface. No other observations were made at any other test substance concentrations. No morphological observations were made. The test substance had no effect on body lengths. Body weights appeared to increase at 100 µg/l.

Under the conditions of this study, the LC50was determined to be 81µg/l, the NOEC was determined to be 32µg/l and the LOEC was determined to be 100µg/L.

 

Short-term toxicity to aquatic invertebrates:

The short-term toxicity of N,N-Didecyl-N-methylpoly(oxyethyl)ammonium Propionate was determined in a 48 hour acute toxicity semi-static study conducted on Daphnia magna.The study was conducted in accordance with Directive 92/69/EEC, Method C2 and OECD Guideline No. 202. 10 daphnids were placed in duplicate, in each test and control vessel at random in the prepared test solutions. The test vessels were covered to reduce evaporation and maintained in at 21°C with a photoperiod of 16 hours light and 8 hours dark. The daphnids were exposed to the test substance at the following concentrations, 0, 0.007, 0.0126, 0.0224, 0.0392, 0.07, 0.126, 0.224, 0.392 and 0.7 mg/L. 100% immobilisation was observed in the 0.7 mg/L and 0.392 mg/L test group after 24 and 48 hour respectively and in the 0.224 mg/L test group after 48 hours. Based on concentration effect relationship observed, the no-observed-effect concentration (NOEC) was found to be 0.0392 mg/L.

 

Long-term toxicity to aquatic invertebrates:

In a study conducted in accordance with OECD Guideline 211, Daphnia magna were exposed to the test material, Bardac 22 with radiolabelled Didecyldimethylammonium Chloride for 21 days, at concentrations of 0, 0.0032, 0.0056, 0.010, 0.018, 0.032, 0.056 mg/l (actual test concentrations, 0, 0.0035, 0.0062, 0.0097, 0.0189, 0.0329, 0.0592 mg/l). The study was conducted in an intermittent flow through system which provided ca. 250 ml of test substance in dilution water every 51 minutes.  Four replicates of each concentration (3.2, 5.6, 10, 18, 32 and 56mg/l) and control media were tested with 10 test animals (<24-hours old).  Under the conditions of this study, the NOEC and LOEC for reproductive effects were determined to be 0.018 mg/l and 0.032 mg/l, respectively. The NOEC and LOEC for survival effects were determined to be 0.010 mg/L and 0.018 mg/L, respectively and for condition effect, the NOEC and LOEC were determined to be 0.018 mg/L and 0.032 mg/L, respectively.

 

Toxicity to aquatic algae and cyanobacteria:

The effect of Bardap 26 (N,N-Didecyl-N-methylpoly(oxyethyl)ammonium Propionate) on the inhibition of algal growth was determined in a 72 hour growth inhibition test on Scenedesmus subspicatus, conducted in accordance with Directive 92/69/EEC, Method C3 and OECD Guideline No. 201. The test substance was dissolved in culture medium and the volume adjusted to 500 ml to give a 200 ml/stock solution from which a series of dilutions were made. The test parameter used was the effect on biomass production and growth rate. The test was conducted at concentrations of 0, 0.0438, 0.0875, 0.175, 0.35 and 0.7 mg/L. Based on concentration effect relationship observed, the no-observed-effect concentration (NOEC) was found to be 0.044 mg/L.

 

Toxicity to microorganisms: 

In a study conducted to determine the effect of Bardap 26 (N,N-Didecyl-N-methylpoly(oxyethyl)ammonium Propionate), activated sludge of a synthetic sewage was exposed to the test material for 3 hours at concentrations of 0, 0.7, 2.24, 7.0, 22.4 and 70 mg/L.

The control group was maintained under identical conditions as treatment groups but not exposed to the test material.

No inhibition on respiration was observed during 3 hours contact time. The test parameter examined was inhibition on respiration rate. Significant inhibition on respiration was observed at 22.4 mg/L after 3 hours contact time. The effect of the test material on the respiration of activated sewage sludge micro-organisms gave a 3-hour EC50 of 16.8 mg/L. Based on concentration effect relationship observed, the no-observed-effect-concentration (NOEC) was found to be 2.24 mg/L.
