- Adsorption and desorption constants:
- Adsorption Freundlich constants of XDE-777 were calculated on seven soils. Between 73.9% and 99.1% of XDE-777 was sorbed to the soils. Adsorption Koc values (unit: mL/g) of XDE-777 ranged from 8961 to 134270 with a geomean of 38217. Adsorption KFoc values (unit: μg1-1/n mL1/ng-1 ) of XDE-777 ranged from 936 to 63394 with a geomean of 5777. The linear correlation coefficients (r2) ranged from 0.898 to 0.995. The 1/n values ranged from 0.608 to 1.066 with an average of 0.819, indicating some concentration dependent sorption behavior.
Desorption Freundlich constants of XDE-777 were calculated on five soils. Less than 20% of the sorbed XDE-777 was desorbed. Desorption Koc values of XDE-777 ranged from 26169 to 142066 with a geomean of 57249. Desorption KFoc values of XDE-777 ranged from 344 to 28871 with a geomean of 3570. The linear correlation coefficients (r2) ranged from 0.694 to 0.988. The 1/n values ranged from 0.517 to 1.037 with an average of 0.755, indicating some concentration dependent sorption behavior.
Adsorption Freundlich constants of X642188 were calculated on nine soils. The percentage of X642188 sorbed to the soil ranged from 45.9% to 92.4%. Adsorption Koc values of X642188 ranged from 2878 to 28825 with a geomean of 6670. Adsorption KFoc values of X642188 ranged from 1626 to 33614 with a geomean of 4518. The linear correlation coefficients (r2) ranged from 0.972 to 0.999 with an average of 0.988. The 1/n values ranged from 0.823 to 1.027 with an average of 0.934 indicating some concentration dependent sorption behavior. - Recovery of test material:
- The average mass balance for XDE-777 Tier 3 adsorption samples was 92.4% ± 4.6% (range 81.9-103.6%). The average mass balance for XDE-777 Tier 3 desorption samples was 97.8 ± 6.4% (range 86.3-111.1%). The average mass balance for X642188 Tier 3 adsorption samples was 97.2 ± 4.2% (range 82.7-104.0%).

No correlation was observed between soil pH and the sorption coefficients of XDE-777 and X642188. - Executive summary:
The adsorption characteristics of XDE-777 were studied in seven soils, Yolo sandy clay (pH 6.9, 0.8% organic carbon) from California/USA, Site K Woodside Farm clay loam (pH 7.2, 4.4% organic carbon) from Rutland/UK, Site J3 Hareby House light clay (pH 7.6, 1.7% organic carbon) from Lincolnshire/UK, Site E1 Farditch farms loam (pH 5.8, 3.9% organic carbon) from Derbyshire/UK, RefSol 03G clay loam (pH5.8, 3.9% organic carbon) from Westphalia/Germany, Montmorenci clay loam (pH 5.5, 1.0% organic carbon) from Indiana/USA, and Fayette clay loam (pH 5.9, 0.9% organic carbon) from Iowa/USA. The desorption characteristics of XDE-777 were studies in the first five soils of the seven soils. The adsorption characteristics of X642188 were determined in Site I2 – Longwoods Quarry sandy loam (pH 7.4, 1.3% organic carbon) from Lincolnshire/UK and Site A1 – UK Acid sand (pH 3.9, 0.8% organic carbon) from Nottinghamshire/UK in addition to the seven soils used in the XDE-777 adsorption. No desorption tests were conducted for X642188.
Soil samples were sterilized by gamma irradiation prior to treatment with test material to minimize microbial degradation during the sorption tests.
The experiment was conducted in accordance with the US EPA Subdivision N. Section 163-1 and Canada PMRA DACO Number 8.2.4.2 - Adsorption/Desorption or OECD Guideline for the Testing of Chemicals –Adsorption/Desorption-106. Sorptive characteristics of the test materials were determined in a batch equilibrium experiment.
The adsorption test of XDE-777 was carried out by equilibrating sterilized soil with the test material at concentrations of 0.15, 0.5, 1.5, 5.0 and 15.0 ng/mL. Equilibration was conducted in the dark at 20°C. The equilibrating solution used was 0.01 M CaCl2, with a soil: solution ratio of 1:25. Due to the rapid degradation of XDE-777 in aqueous solutions, samples were equilibrated only for 0.5 – 2 hours. The desorption phase of XDE-777 was carried out by equilibrating the samples in the dark at 20 °C. The samples were desorbed once, equilibrating for 0.5-2 hours depending on the soil types depending on the soil type.
The adsorption test of X642188 was carried out by equilibrating sterilized soil with the test material at concentrations of 2, 10, 20, 100 and 200 ng/mL. Equilibration was conducted in the dark at 20 °C. The equilibrating solution used was 0.01 M CaCl2, with a soil: solution ratio of 1:25. Due to the rapid degradation of X642188 in aqueous solutions, samples were equilibrated only for 4 hours only.
Soil and aqueous phases were separated by centrifugation after the adsorption or desorption steps. Soils samples were extracted three times with 80:20 acetonitrile: water containing 0.5% formic acid, centrifuged, and the extracts decanted and combined. XDE-777 and X642188 residues in aqueous solution and organic extracts were assayed by both LSC and LCMS. 14C residue remaining in the soil pellet after extraction was determined by oxidative combustion. Adsorption and desorption parameters were calculated using the Freundlich adsorption and desorption isotherms. Only Freundlich adsorption isotherms were calculated for X642188.
