Skin irritation

The dermal irritation potential of test article was determined according to the OECD 439 In Vitro Skin Irritation: Reconstructed Human Epidermis Test Method”. The MatTek EpiDerm™ model was used to assess the potential dermal irritation of the test article by determining the viability of the tissues following exposure to the test article via MTT. The objective of this study was to assess the dermal irritation potential of test article Tissues were exposed to test article and controls for ~one hour, followed by a 42 hour post-exposure recovery period. The viability of each tissue was determined by MTT assay. 

The MTT data shows that the assay quality controls were met. The mean tissue viabilities for the Positive control, Methyl acetate were 6.5%, 10.7% respectively in the first and second run, whereas the tissue viabilities of the negative control, Tissue culture water remained at 100% in the both the runs.

ForSodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl] benzoxazolesulphonate, CAS No. 93859-32-6, from the three test-article treated tissues, the individual viabilities were 78.4%, 69.2% and 107.1% with a mean tissue viability of 84.9%.  The protocol states that the standard deviation of the three treated tissues not be greater than 18%. The test-article treated tissues had a standard deviation of 19.79%. Although the standard deviation did not meet the acceptance criterion, all the individual tissue viabilities were above the classification cut-off of 50%. Therefore the material will be classified as Non-Irritating. 

The Mean % tissue viability compared to negative control (n=3) of Sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl] benzoxazolesulphonate[CAS: 93859-32-6] was determined to be 84.9%.

Hence, under the experimental test conditions it was concluded that Sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl] benzoxazolesulphonate[CAS: 93859-32-6] was considered to be not irritating to the human skin and being classified as “Not Classified'' as per CLP Regulation.

An ocular irritation potential of target chemical sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) was assessedin various experimental studies which were conducted on rabbits for target chemicalsodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) and its structurally similar read across substancesIndigo carmine (CAS No: 860-22-0) and Patent Blue V (CAS No: 129-17-9). The predicted data usingQSAR toolboxhas also been compared with the experimental data.Based on the available data for the target and read across substances and applying the weight of evidence approach, it can be concluded that chemical sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) is unable to cause eye damage and thus can be considered as not irritating. Comparing the above annotations with the criteria of CLP regulation, it can be classified under the category “Not Classified”.

EpiDerm™ Tissue SamplesEpiDerm™ tissues, Lot No. 27646 Kits I and J, were received from MatTek on 12 Dec 2017, and Lot No. 27654 Kits O and P, were received from MatTek on 19 Dec 2017 All tissues were refrigerated at 2-8°C upon receipt. These test articles were assessed for compatibility with pre-cut nylon mesh supplied with the tissues. The mesh was placed on a slide and 30 μl of a liquid test articles or PBS (negative control) were applied. After 60 minutes of exposure, the mesh was checked microscopically. If no damage or other interaction was observed, indicating that the mesh was compatible with the test article, the mesh was used as a spreading aid.Tissue Viability (MTT Reduction)At the end of the incubation period, each EpiDerm™ tissue was rinsed with PBS and transferred to a 24-well plate containing 300 μl of MTT solution (1 mg/ml MTT in DMEM). Following the MTT incubation period, each EpiDerm™ tissue was rinsed with PBS and then treated with 2.0 ml of extractant solution (isopropanol) per well for at least two hours, with shaking, at room temperature. Two aliquots of the extracted MTT formazan were measured at 540 nm using a plate reader (μQuant Plate Reader, Bio-Tek Instruments, Winooski, VT).For several tissues, the test article had stained the tissues. Therefore, the tissues were extracted with only 1.0 ml, allowing extraction to occur only through the bottom of the insert. After the extraction period, the tissue insert was removed and discarded and 1.0 ml of extraction solution were added to each well, bringing the volume to a total of 2.0 ml.Quality ControlsThe assay meets the acceptance criteria if the mean OD540 of the negative control tissues is between 1.0 and 2.5, inclusive, and the mean viability of positive control tissues, expressed as percentage of the negative control tissues, is at least 20%. Various studies has been investigated for the test chemical sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) to observe the potential for drmal irritation to a greater or lesser extent. The studies are based on in vitro and in vivo experiments in rabbits for target chemical sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) and its structurally similar read across substances Aluminium, 2-(1,3-dihydro-3-oxo-5-sulfo-2H-indol-2-ylidene)-2,3-dihydro-3-oxo-1H-indole-5-sulfonic acid complex (CAS No. 16521-38-3) and Disodium 5,5’-(2-(1,3-dihydro-3-oxo-2H-indazol-2-ylidene)-1,2-dihydro-3H-indol-3-one)disulphonate (CAS No. 860-22-0) .

 

The dermal irritation potential of test article was determined according to the OECD 439 In Vitro Skin Irritation: Reconstructed Human Epidermis Test Method”. The MatTek EpiDerm™ model was used to assess the potential dermal irritation of the test article by determining the viability of the tissues following exposure to the test article via MTT. The objective of this study was to assess the dermal irritation potential of test article Tissues were exposed to test article and controls for ~one hour, followed by a 42 hour post-exposure recovery period. The viability of each tissue was determined by MTT assay. 

The MTT data shows that the assay quality controls were met. The mean tissue viabilities for the Positive control, Methyl acetate were 6.5%, 10.7% respectively in the first and second run, whereas the tissue viabilities of the negative control, Tissue culture water remained at 100% in the both the runs.

ForSodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl] benzoxazolesulphonate, CAS No. 93859-32-6, from the three test-article treated tissues, the individual viabilities were 78.4%, 69.2% and 107.1% with a mean tissue viability of 84.9%.  The protocol states that the standard deviation of the three treated tissues not be greater than 18%. The test-article treated tissues had a standard deviation of 19.79%. Although the standard deviation did not meet the acceptance criterion, all the individual tissue viabilities were above the classification cut-off of 50%. Therefore the material will be classified as Non-Irritating. 

The Mean % tissue viability compared to negative control (n=3) of Sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl] benzoxazolesulphonate[CAS: 93859-32-6] was determined to be 84.9%.

Hence, under the experimental test conditions it was concluded that Sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl] benzoxazolesulphonate[CAS: 93859-32-6] was considered to be not irritating to the human skin and being classified as “Not Classified'' as per CLP Regulation.

 The above result was supported by two studies designed and conducted by Sustainability Support Services (Europe) AB, 2017 to determine the dermal reaction profile of read across chemicals Aluminium, 2-(1,3-dihydro-3-oxo-5-sulfo-2H-indol-2-ylidene)-2,3-dihydro-3-oxo-1H-indole-5-sulfonic acid complex (CAS No. 16521-38-3) and Disodium 5,5’-(2-(1,3-dihydro-3-oxo-2H-indazol-2-ylidene)-1,2-dihydro-3H-indol-3-one)disulphonate (CAS No. 860-22-0) in Sprague Dawley rats. These studies were performed as per OECD Guidelines 402 and complying to the GLP procedures. In both the studies, the test item was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days.    Animals exhibited normal body weight gain through the study period of 14 days. Gross pathological examination did not reveal any abnormalities attributable to the treatment. The overall irritation score of the substance was obtained to be 0 and no erythema and edema (skin irritation) were observed at the end of 14 days observation period after patch removal. Hence, it was concluded that the substances Aluminium, 2-(1,3-dihydro-3-oxo-5-sulfo-2H-indol-2-ylidene)-2,3-dihydro-3-oxo-1H-indole-5-sulfonic acid complex (CAS No. 16521-38-3) and Disodium 5,5’-(2-(1,3-dihydro-3-oxo-2H-indazol-2-ylidene)-1,2-dihydro-3H-indol-3-one)disulphonate (CAS No. 860-22-0) were non-Irritating to the skin of rats under the experimental conditions tested .

 

Based on the available data for the target and read across substances, it can be concluded that chemical sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) is unable to cause skin irritation and considered as not irritating. In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, ocular irritation potential was estimated for sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6). The substance sodium 5-(aminosulphonyl)-2-[7-(diethylamino)-2-oxo-2H-1-benzopyran-3-yl]benzoxazolesulphonate (CAS No: 93859-32-6) is considered to be not irritating to the eyes of rabbits.

 

The EFSA FEEDAP Panel (EFSA Panel on Additives and Products or Substances used in Animal Feed (FEEDAP), European Food Safety Authority (EFSA), Parma, Italy; EFSA Journal 2015; 13(5):4108) conducted an ocular irritation study of read across chemical Indigo carmine (CAS No: 860-22-0) in rabbits by using Draize method. In the given study, when 0.2mL of 10% aqueous solution was applied twice daily for five days per week for four weeks to the eye of six rabbits slight transient irritation was observed. Three days after the last administration, there were no signs of eye irritation in any of the treated animals. Hence the Indigo carmine chemical (CAS No: 860-22-0) was considered to be not irritating to the rabbits’ eye.

 

The above results were further supported by the experimental study reported by COMMISSION OF THE EUROPEAN COMMUNITIES (Scientific Committee on Cosmetology (seventh series), 1988) for read cross chemical Patent Blue V (CAS No: 129-17-9) in rabbits. During the study, 0.25 ml of a saturated solution of the test chemical in saline was instilled into the eyes of rabbits. The chemical did not induce any signs of ocular lesions. Hence the chemical Patent Blue V (CAS No: 129-17-9) was considered to be not irritating to the rabbits’ eye.
