The studies are based on in vivo experiments in rabbits and guinea pigs for target chemicalSodium tris(1,2-naphthoquinone 1-oximato-O,O')ferrate(1-) (CAS No: 16143-80-9) and its closely related read across substances Benzyl cinnamate (CAS No: 103-41-3)andBenzyl butyl phthalate (CAS No: 85-68-7).The predicted data using the OECD QSAR toolbox has also been compared with the experimental data and summarized as below;

 

The COMMISSION OF THEEUROPEAN COMMUNITIES reported skin irritation study for target chemicalSodium tris(1,2-naphthoquinone 1-oximato-O,O')ferrate(1-) (CAS No: 16143-80-9) in rabbits. When 500 mg of 50% aqueous suspension or 500mg (0.5g) of undiluted Sodium tris(1,2-naphthoquinone 1-oximato-O,O')ferrate(1-) was applied on intact or abraded skin of each rabbit, none of the treated rabbits showed positive skin reaction. The Primary Skin irritation study was conducted by D.L.J. Opdyke (Food and Cosmetics Toxicology 1979, Pages 132–133) for read across chemical Benzyl cinnamate (CAS No: 103-41-3) on 25 human subjects that supported the abov mentioned results for target chemical. Each subject was treated with 8% of Benzyl cinnamate (CAS No: 103-41-3) in petrolatum dermally for 48 hours under a closed path patch.Since no known signs of irritation was observed, the chemicalBenzyl cinnamate (CAS No: 103-41-3)was considered to be not irritating to theskin of human subjects.

 

The above results were supported by dermal irritation study conducted by Cosmetics Ingredient Review Panel (International Journal of Toxicology,Vol 11, Issue 1, pp. 1 – 23, 1992) of read across chemical Benzyl butyl phthalate (CAS No: 85-68-7) in 6 New Zealand White rabbits. Undiluted Benzyl Butyl Phthlate, 0.5 ml, was applied to the intact and abraded skin of rabbits and effects were observed for 24 h. There was practically no irritation to the rabbit skin exposed to Benzyl butyl phthalate for 24 h. Hence, Benzyl butyl phthalate can be considered as not irritating to New Zealand White rabbit skin.
