Skin Irritation:
Several studies were performed to determine the degree of dermal irritation caused by Trisodium 2-[(E)-2-(3-methyl-5-oxo-1-{4-[2-(sulfonatooxy)ethanesulfonyl]phenyl}-4,5-dihydro-1H-pyrazol-4-yl)diazen-1-yl]naphthalene-1,5-disulfonate in living organisms. These studies include in vivo studies in rabbits as well as predicted data for the target chemical as well its structurally similar read across substances,Tetrasodium 1-acetamido-2-hydroxy-3-(4-((4-sulphonatophenylazo)-7-sulphonato-1-naphthylazo))naphthalene-4,6-disulphonate(CI Food Black 1)[CAS: 2519-30-4], 1-(2-methoxy-5-methyl-4-sulfophenyl-1-azo)-6 sulfo-2-hydroxynapthalene,disodium salt ( ALLURA RED AC)[CAS: 25956-17-6] and Tetrasodium (3E)-6-amino-4 -oxo-3-(2-{7-sulfinato-4-[(E)-2-(4-sulfonatophenyl)diazen-1-yl]naphthalen-1-yl}hydrazin-1-ylidene)-3,4-dihydronaphthalene-2,7-disulfonate(CI Food Black 2)[CAS: 2118-39-0]. Thepredicted data using the OECD QSAR toolbox has also been compared with the experimental data.
In a prediction done by SSS (2017) using the OECD QSAR toolbox v3.3 with log kow as the primary descriptor, the dermal irritation potential was estimated forTrisodium 2-[(E)-2-(3-methyl-5-oxo-1-{4-[2-(sulfonatooxy)ethanesulfonyl]phenyl}-4,5-dihydro-1H-pyrazol-4-yl)diazen-1-yl]naphthalene-1,5-disulfonate. Trisodium 2-[(E)-2-(3-methyl-5-oxo-1-{4-[2-(sulfonatooxy)ethanesulfonyl]phenyl}-4,5-dihydro-1H-pyrazol-4-yl)diazen-1-yl]naphthalene-1,5-disulfonate was estimated to be not irritating to the skin of New Zealand White rabbits.
This result is supported by the experimental study conducted(Sustainability Support Services (Europe) AB, 2016) to determine the dermal reaction profile of the structurally similar read across substance, Tetra sodium 1-acetamido-2-hydroxy-3-(4-((4-sulphonatophenylazo)-7-sulphonato-1-naphthylazo))naphthalene-4,6-disulphonate[CAS:2519 -30 -4] in Sprague Dawley rats. The study was performed as per OECD Guidelines 402 and complying to the GLP procedures. Ten rats (5 male and 5 female) were used for conducting dermal irritation /corrosion study.
The animals were kept in their cages for at least 5 days prior to administration for acclimatization to the laboratory condition and after acclimatization period, animals were randomly selected. Approximately 24 hours before application, the hair of each rat was closely clipped from the trunk (dorsal surface and sides from scapular to pelvic area) with an electric clipper, so as to expose at least 10% of the body surface area. The test item was moistened with distilled water. The test item was applied onto the exposed skin of the animal, taking care to spread the test item evenly over the entire area of approximately 10% of the total body surface area or as much of the area as can reasonably be covered. The test item was held in contact with the skin using a porous gauze dressing and non irritating tape around the animal to cover the exposure site for first 24 hours exposure period. Elizabethan collar was placed on each animal for first 24 hours after application of the test item. These collars prevent ingestion of test item. Following 24 hours of exposure, the wrapping was removed and the test site wiped free of excess test item. Distilled water was used to remove residual test item.
The test item Tetra sodium 1-acetamido-2-hydroxy-3-(4-((4-sulphonatophenylazo)-7-sulphonato-1-naphthylazo))naphthalene-4,6-disulphonate was applied to shorn skin of 5 male and 5 female animals at 2000 mg/kg body weight. Administration of the test item at 2000 mg/kg did not result in any skin reaction at the site of application during the study period of 14 days. Administration of the test item did not result in any signs of toxicity and mortality during the study period of 14 days. Animals exhibited normal body weight gain through the study period of 14 days. Gross pathological examination did not reveal any abnormalities attributable to the treatment.
The overall irritation score of the substance was determined to be 0 and no erythema and edema (skin irritation) were found at the end of 14 days observation period after patch removal.
Hence, it was concluded that Tetra sodium 1-acetamido-2-hydroxy-3-(4-((4-sulphonatophenylazo)-7-sulphonato-1-naphthylazo))naphthalene-4,6-disulphonate was Non-Irritating to the skin of Sprague Dawley rats under the experimental conditions tested and classified as “Category- Not Classified” as per CLP Classification.
The above studies are supported by the experimental study summarized in HSDB (Hazardous Substances Data Bank)for allura red acid dye AC.U.S National Library of Medicine ; 2011; for the structurally similar read across substance, 1-(2-methoxy-5-methyl-4-sulfophenyl-1-azo)-6 sulfo-2-hydroxynapthalene,disodium salt ( ALLURA RED AC)[CAS:25956-17-6]. 25% neat or aqueous solution of Disodium 6-hydroxy-5-[(2-methoxy-4-sulphonato-m-tolyl)azo]naphthalene-2-sulphonate was applied to the skin of 200 human volunteers for 72 hours. This was followed by one 48 hours application 10-14 days. After the exposure, the human volunteers were observed for signs of irritation. None of the subjects exhibited compound induced irritation. Hence, disodium 6-hydroxy-5-[(2-methoxy-4-sulphonato-m-tolyl)azo]naphthalene-2-sulphonate was considered to be not irritating to human skin.
These studies are further supported by the experimental study summarized in Scientific Committee on Cosmetology (seventh series), 1988; for the structurally similar read across substance, Tetrasodium (3E)-6-amino-4-oxo-3-(2-{7-sulfinato-4-[(E)-2-(4-sulfonatophenyl)diazen-1-yl]naphthalen-1-yl}hydrazin-1-ylidene)-3,4-dihydronaphthalene-2,7-disulfonate(CI Food Black 2)[CAS: 2118-39-0]. Rabbits were given intracutenous injection of 0.1 ml 5% solution of the test chemical on their back skin and observed for signs of dermal reactions (exact duration of study not mentioned). The intracutenous injection did not induce any cutenous damage to the skin. A black spot formed on the on back skin disappeared largely during the second day and was completely invisible after 2 days. Hence, Food Black 2 can be considered to be not irritating to skin.
Available studies for the target and structurally similar read across substances indicate a very strong possibility ofTrisodium 2-[(E)-2-(3-methyl-5-oxo-1-{4-[2-(sulfonatooxy)ethanesulfonyl]phenyl}-4,5-dihydro-1H-pyrazol-4-yl)diazen-1-yl]naphthalene-1,5-disulfonatebeing not irritating to skin. Hence,Trisodium 2-[(E)-2-(3-methyl-5-oxo-1-{4-[2-(sulfonatooxy)ethanesulfonyl]phenyl}-4,5-dihydro-1H-pyrazol-4-yl)diazen-1-yl]naphthalene-1,5-disulfonatecan be considered to be not a skin irritant. A study was performed by S.D. Gettings et.al, 1992 to determine the ocular irritation potential of thestructurally similar read across substance,trisodium 5-hydroxy-1-(4-sulphophenyl)-4-(4 -sulphophenylazo)pyrazole-3-carboxylate(Tartrazine)[CAS: 1934-21-0].The study was performed according to a modification of the Draize test (Draize, 1959).
Tartrazine (FD & C Yellow No. 5) was prepared daily as a 3% (w/v) suspension in aqueous vehicle containing 0.5% (w/v) hydroxypropyl methylcellulose and 0.25% (w/v) laureth -10 acetate. Tartrazine (FD & C Yellow No. 5) 3% w/v in aqueous vehicle was administered once daily, for a total of 21 days, to the conjunctival sac of the right eye of New Zealand White Rabbits (6 of each sex/ group) at a dose volume of 30µl. Control animals (6 of each sex) received 30/µl of the vehicle daily. Ocular irritation was determined according to a modification of the Draize test (Draize, 1959). Interpretation of observations and assignment of scores were consistent with those described by the Consumer Product Safety Commission (1972).All eyes were scored for ocular irritation pretest (8 days, 24 hr and immediately prior to the initial dose) and approximately 24 hr after each treatment, prior to the next instillation of test material; on days 1, 3, 7, 14 and 21, the eyes were also evaluated for irritation 1 hr after treatment. In addition, all readily observable ocular structures were evaluated for eye stain and particle embedment 24 hr after each treatment.
No lethality or significant clinical signs, and no substance-related weight change were observed. Except slight conjunctival redness or discharge, that were seen sporadically in the eyes of the animals, all animals were free of significant signs of ocular irritation. No significant signs of eye staining or particle depositions were observed. At ophthalmoscopic examinations, no ocular abnormities were noticed.
Hence, FD&C Yellow No.5 (tartrazine) was not expected to cause any irritant effects following repeated exposure to rabbit eyes.
These results are supported by the experimental study summarized in Environment and Quality of Life - Reports (Seventh Series) , European Commission (EC) - Scientific Committee on Cosmetology (SCC) , 1988; for the structurally similar read across substance, 1-(2-methoxy-5-methyl-4-sulfophenyl-1-azo)-6 sulfo-2-hydroxynapthalene,disodium salt ( ALLURA RED AC)[CAS: 25956-17-6]. Disodium 6-hydroxy-5-[(2-methoxy-4-sulphonato-m-tolyl)azo]naphthalene-2-sulphonate was subjected to rabbit eye in the concentration 0.1 ml1.0% aqueous solution .No ocular reaction was observed .Hence , Disodium 6-hydroxy-5-[(2-methoxy-4-sulphonato-m-tolyl)azo]naphthalene-2-sulphonate was considered to be not eye irritant.
These studies are further supported by the experimental study summarized in Scientific Committee on Cosmetology (seventh series), 1988; for the structurally similar read across substance, Tetrasodium (3E)-6-amino-4-oxo-3-(2-{7-sulfinato-4-[(E)-2-(4-sulfonatophenyl)diazen-1-yl]naphthalen-1-yl}hydrazin-1-ylidene)-3,4-dihydronaphthalene-2,7-disulfonate(CI Food Black 2)[CAS: 2118-39-0]. Undiluted 50 mg of test chemical was instilled in the conjunctival sac of 2 rabbits for 24 hours.
Slight erythema was observed in the rabbit eyes which disappeared in 24 hours. No corneal damage was observed. Hence, Food Black 2 can be considered to be not irritating to eyes.
Available studies for the target and structurally similar read across substances indicate a very strong possibility ofTrisodium 2-[(E)-2-(3-methyl-5-oxo-1-{4-[2-(sulfonatooxy)ethanesulfonyl]phenyl}-4,5-dihydro-1H-pyrazol-4-yl)diazen-1-yl]naphthalene-1,5-disulfonatebeing not irritating to eyes.
