Details on sampling:
- Concentrations: 6.25, 12.5, 25.0, 50.0 and 100.0 mg/L
- Sampling method: The samples of fresh solutions at test initiation and samples of 24 old hour solution at first renewal
(after 24 hours) collected from each test concentration and the control were analysed. The samples of
fresh solution at renewal and 24 hour old solution at renewal and at test termination collected from
only the highest and the lowest test concentrations and the control were analysed.
Vehicle:

Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- The saturated solution was prepared by 24 hour slow stirring of the test substance at a concentration of 100 mg/L in a closed volumetric bottle of 5 L capacity with small headspace at temperature 25°C. The saturated solution was left to stand for 1 hour to let undissolved test substance accumulate on the surface. The saturated solution was taken from the bottom of the test vessel by sampling tube and was thoroughly checked for undissolved test material and turbidity. The transmittance of the basic solution was from 94.4 to 98.7% measured for the fresh solutions . Any substance from the surface film was taken because the solution was taken from the bottom of the bottle. The saturated solution
was diluted by a factor of 2 to obtain the lower test concentrations.
Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: Daphnia magna Straus
- Source: Institute of Industrial Organic Chemistry, Branch Pszczyna, Department of Ecotoxicology, Laboratory of Aquatic Toxicology [SOP/W/67]
- Age at study initiation (mean and range, SD): < 24 h old, not first brood progeny, range of parent daphnid age: 21 – 25 days, neonates collected
from laboratory culture in the Institute of Industrial Organic Chemistry.
- Feeding during test: no feeding
- Method of breeding: Daphnia magna was cultured in glass beakers of 150 mL volume (one parent per vessel) at room
temperature 18 – 22ºC with the light : dark ratio of 16 h : 8 h The culture was maintained in Elendt M7 medium
- Food type: Pseudokirchneriella subcapitata algae suspension originating from the culture at the
Laboratory of Aquatic Toxicology [SOP/W/65]. Group B vitamins and micronutrients necessary for proper growth were supplied with lyophilized Spirulina sp. suspension.

TEST SYSTEM
- Test vessel: glass bottles of 100 mL , with glass caps.
- Type (delete if not applicable): closed
- Aeration: no
- Renewal rate of test solution (frequency/flow rate): 24 h (due to volatility a decrease of the content of the test item in water)

TEST MEDIUM / WATER PARAMETERS
-The Elendt M7 medium recommended by OECD Guideline No 202 (2004) was used for culturing and as the diluent/solvent for the test item.

OTHER TEST CONDITIONS

- Photoperiod: day: night regime 16 : 8 h

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) :
- Daphnids were observed for immobilization after 24 and 48 hours of exposure. The test organisms were considered immobile if they showed no ability to swim within 15 seconds after swirling the test vessel.
-The temperature of the solutions was constantly recorded in an additional test vessel with test medium adjancent to the test vessels.
-The pH and dissolved oxygen content were measured at test initiation before division into replicates and at test termination in pooled replicates .
