- Concentrations: The control and loading rates of 0.32, 1.0, 3.2, 10, 32 and 100 mg/L were analysed analytically. The analytical samples of the lowest concentration with the loading rate 0.10 mg/L were not analyzed as this loading rate was below the NOEC determined in this test and thus not relevant for the interpretation of the biological results.
- Sampling method: Duplicate samples without algae were taken from each loading rate and the control at the start of the test. At the end of the test (after 72 h), samples (containing algae) were taken in duplicate from all loading rates and from the control. For sampling from the aged test media, the contents of the respective replicates were combined prior to sampling.

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Water Accommodated Fractions (WAFs) with individual dispersions of the test item with the loading rates of 1.0, 3.2, 10, 32 and 100 mg/L were prepared. Thereafter the dispersions were stirred for 3 hours to dissolve the different components of the test item as far as possible within a reasonable time period. After stirring, the dispersions were let to settle for 30 minutes and thereafter the lower phases were filtered through a 0.45 µm membrane filter. The filter was saturated by at least 200 mL test medium and the negative pressure of the filtration unit was reduced to a minimum to avoid losses of the volatile test item. These undiluted filtrates were tested as WAFs. Due to technical reasons (operational constraints on the precision of direct pipetting of test item), the WAFs with the lower loading rates of 0.32 mg/L and 0.10 mg/L were prepared as dilutions of the WAF with the loading rate of 1.0 mg/L.

- Exponential growth in the control (for algal test): yes
- Observation of abnormalities (for algal test): The microscopic examination of the algal cells at the highest test concentration of 100 mg/L at the end of the test showed a disfiguration of most of the algal cells as compared to the control. However, no difference was determined between the algae growing at the loading rates of 32 and 10 mg/L and the algal cells in the control.
- Any observations (e.g. precipitation) that might cause a difference between measured and nominal values: No remarkable observations were made concerning the appearance of the test media. All test media were clear solutions throughout the test period.

Reported statistics and error estimates:
The 72-hour EL10, EL20 and EL50 values for the inhibition of average growth rate and yield and their 95% confidence intervals were calculated by Probit Analysis using linear maximum likelihood regression. For the determination of the LOEL and NOEL, the average growth rate and yield at the test concentrations were compared to the control values by Williams t-test and Welch t test after Bonferroni-Holm-adjustment, where appropriate. Statistical analysis was performed using ToxRat Professional®.

Additional information

One experimental study is available investigating the toxicity of the test item to aquatic algae. The impact of the test item on the growth of the freshwater green algal species Pseudokirchneriella subcapitata was investigated in a 72-hour test according to OECD 201 (2011) under GLP conditions. Since the test item is a complex mixture consisting of several constituents with differently low solubility, Water Accommodated Fractions (WAFs) with the loading rates of 0.10, 0.32, 1.0, 3.2, 10, 32 and 100 mg/L were tested to assess the toxicity of the test item to algae. A control group (test water without test item) was tested in parallel. For preparation of the WAFs, individual dispersions of the test item with the loading rates of 1.0, 3.2, 10, 32 and 100 mg/L were prepared. Thereafter the dispersions were stirred for 3 hours to dissolve the different components of the test item as far as possible within a reasonable time period. After stirring, the dispersions were let to settle for 30 minutes and thereafter the lower phases were filtered through a 0.45 µm membrane filter. These undiluted filtrates were tested as WAFs. Due to technical reasons (operational constraints on the precision of direct pipetting of test item), the WAFs with the lower loading rates of 0.32 mg/L and 0.10 mg/L were prepared as dilutions of the WAF with the loading rate of 1.0 mg/L. The test item gave a chromatographic profile consisting of several peaks. The analytical measurements of the soluble components were based on the sum of these peaks. At the start of the test, the measured concentrations of the analyzed components of the test item in the test media with loading rates of 0.32, 1.0, 3.2, 10, 32 and 100 mg/L were 0.24, 0.71, 1.9, 5.1, 11 and 19 mg/L, respectively. The measured concentrations at the end of the test were in the range of 56 and 96 % of the initially measured values. The 72-hour EL10 and EL50 values for the inhibition of growth rate were found to be 52 and 11 mg/L, respectively. Since WAFs were tested, all effect values were based on the nominal loading rates of the test item. The preparation of the WAFs and procedure of evaluation of data of WAFs is based on recommendations of the OECD Guidance Document No. 23 on Aqueous-Phase Aquatic Toxicity Testing of Difficult Test Chemicals, 2019.
